摘 要:CHX(Cation/H+ exchanger)基因家族是CPA(Cation proton antiporter)基因家族的一个亚家族,家族基因广泛参与维持植物体内Na+/K+离子平衡的调控过程,对植物维持细胞离子浓度、花器官的发育起着至关重要的作用。本研究以葡萄为材料,共鉴定出19个VvCHX基因,命名为VvCHX1~VvCHX19。运用生物信息学的方法,系统地分析了葡萄VvCHXs基因的结构、蛋白理化性质、保守基序、系统进化,同源基因分析和组织特异性表达。结果表明,根据多物种CHX蛋白的进化树分析将19个Vv CHX家族成员划分为4类;VvCHX家族基因不均匀的分布于7条染色体上,其中3对基因存在串联重复现象;同源基因分析表明,葡萄VvCHX家族成员可能在维持离子浓度、花器官发育中起到重要作用;组织特异性表达分析表明,绝大多数葡萄VvCHX基因家族成员在雄蕊、花粉、花中表达量较高。本研究进一步探究CHX基因家族调控植物抗逆的分子机制提供依据。The CHX(Cation/H^+ exchanger) gene family is a subfamily of the CPA(Cation proton antiporter) gene family. The CHX family genes are widely involved in the regulation of Na^+/K^+ ion balance in plants, and play a vital role in maintaining plant ion concentration and flower organ development. In this study, 19 VvCHX genes,designated VvCHX1 to VvCHX19, were identified by the grape genome. The gene structure, protein physicochemical properties, conserved motifs, phylogenetic tree analysis, homologous gene analysis and tissue-specific expression of Vv CHXs genes were analyzed by bioinformatics approaches. The results showed that 19 VvCHX family members were classified into 4 categories according to the phylogenetic tree analysis of multi-species CHX proteins. The VvCHX family members were found to be unevenly distributed on 7 chromosomes, and three pairs of genes were found to have tandem duplication. According to homologous gene analysis, the members of VvCHX family may play an important role in maintaining ion concentration and flower organ development. Tissue-specific expression analysis indicated that most grape VvCHX gene family members expressed higher levels in stamens,pollen and flowers. The study laid the foundation for exploring the molecular mechanism of the CHX gene family regulating plant stress resistance.
