摘  要:为通过组织培养的方法对喜马拉雅樱花(Cerasus cerasoides D. DON)进行快速规模化繁殖,本研究以喜马拉雅樱花带腋芽的幼嫩茎段为外植体,研究不同浓度的细胞分裂素对腋芽启动的影响,然后采用正交试验设计方法对不定芽增殖培养基和生根培养基的组分进行优化,同时研究了不同栽培基质对炼苗的影响。实验结果表明,添加6-苄氨基嘌呤(6-BA) 0.2 mg/L的MS培养基最适合诱导腋芽的萌发。适合不定芽增殖的培养基为WPM,同时添加0.5 mg/L 6-BA,增殖系数为5.5倍左右。适合组培苗生根的培养基组分为1/2MS,添加1.0 mg/L吲哚丁酸(IBA)、1.0 mg/L萘乙酸(NAA)、0.4 g/L活性炭。在草炭∶珍珠岩=1∶1的基质上,组培苗的移栽成活率最高。本研究建立了一种利用嫩茎进行离体快速繁殖的方法,可应用于喜马拉雅樱花种苗的生产。In order to propagate Himalayan cherry(Cerasus cerasoides D.DON) rapidly through tissue culture, we used young stem nodes with axillary buds from Himalayan cherry as explants, and researched the effect of different concentration of cytokinin on axillary buds initiation. Then the medium components for shoots proliferation and plant rooting were improved by orthogonal experimental design and the effects of different substrates on survival rate of transplantation were investigated. As a result, axillary buds were initiated on the MS medium, supplemented with 0.2 mg/L 6-Benzylaminopurine(6-BA). The medium suitable for adventitious bud proliferation was WPM, and 0.5 mg/L 6-BA was added at the same time, with a proliferation coefficient of about 5.5 times. The 1/2 MS medium containing 1.0 mg/L 3-indolebutyric acid(IBA), 1.0 mg/L 1-naphthylacetic acid(NAA) and 0.4 g/L activated carbon was suitable for shoots rooting. Plantlets get the highest survival rate when transplanted onto the substrate of Peat ∶Perlite =1 ∶1. We established a micropropagation method for Himalayan cherry by using young stem nodes,which would benefit seedling production.
