摘 要:目的:研究活动期系统性红斑狼疮(SLE)患者外周血中白介素-38(IL-38)和IKKβ激酶的表达水平,以探讨IL-38与IKKβ的关系.方法:采用实时荧光定量聚合酶链反应检测IL-38、IKK基因表达水平;酶联免疫吸附试验(ELISA)检测血清IL-38、肿瘤坏死因子α(TNF-α)蛋白表达水平;采用t检验或Mann-Whitney秩和检验.结果:①活动期SLE患者IL-38 mRNA表达水平(0.36±0.09)显著低于正常对照组(1.00±0.17),差异有统计学意义(P<0.01);活动期SLE患者IL-38蛋白表达水平(5.86±2.76)pg/mL显著低于正常对照组(18.48±1.35)pg/mL,差异有统计学意义(P<0.05);②活动期SLE患者TNF-α表达水平(10.78±1.25)pg/mL显著高于正常对照组(4.34±0.69)pg/mL,差异有统计学意义(P<0.05);③活动期SLE患者IKKβmRNA表达水平(6.01±1.51)显著高于正常对照组(1.16±0.14),差异有统计学意义(P<0.05);④活动期SLE患者IL-38与TNF-α蛋白表达水平呈负相关,差异有统计学意义(P<0.05);⑤活动期SLE患者IL-38与IKKβmRNA表达水平呈负相关,差异有统计学意义(P<0.05).结论:活动期SLE患者IL-38基因及蛋白表达水平下降,并且与TNF-α和IKKβ水平呈负相关,推测在SLE患者体内IL-38水平下降,促进TNF-α和IKKβ的高表达,从而活化核因子-κB(NF-κB)信号通路,参与了SLE的发病.Objective:To study the expression of interleukin-38(IL-38)and IKKβkinase in peripheral blood of patients with active systemic lupus erythematosus(SLE),and to explore the relationship between IL-38 and IKKβ.Methods:The gene expression levels of IL-38 and IκB kinase complex were detected by real-time fluorescence quantitative polymerase chain reaction(PCR),serum IL-38 and tumor necrosis factor-α(TNF-α)levels were detected by enzyme-linked immunosorbent assay(ELISA),and t-test or Mann-Whitney rank sum test were used.Results:The expression level of IL-38 mRNA in active SLE patients(0.36±0.09)was significantly lower than that in the normal control group(1.00±0.17),and the difference was statistically significant(P<0.01).The expression level of IL-38 protein in active SLE patients(5.86±2.76)was significantly reduced as compared with normal control group(18.48±1.35),and the difference was statistically significant(P<0.05).Expression of TNF-αin patients with active SLE(10.78±1.25)was significantly higher than that in the normal control group(4.34±0.69),the difference was statistically significant(P<0.05).The expression level of IKKβmRNA(6.01±1.51)in active SLE patients was significantly higher than that in normal control group(1.16±0.14),the difference was statistically significant(P<0.05).There was a negative correlation between the expression of IL-38 and TNF-α(P<0.05)and between the expression of IL-38 mRNA and IKKβmRNA(P<0.05)in patients with active SLE.Conclusion:The level of IL-38 gene and protein expression in active SLE patients was decreased,and negatively correlated with the level of TNF-αand IKKβ.It is speculated that the decrease of IL-38 level in SLE patients can promote the high expression of TNFαand IKK,thus causing the activation of NF-κB signaling pathway,which is involved in the pathogenesis of SLE.
