摘  要:目的:探讨十二指肠黏膜机械屏障在功能性消化不良发病的影响及电针治疗功能性消化不良的作用机制。方法:将30只SD大鼠按随机数字表法分为空白对照组、模型组、电针组各10只,模型组及电针组采用多因素干预法造模20 d,空白对照组不给予任何处置,造模成功后电针组针刺“后三里”穴、“太冲”穴后接电针治疗14 d,余组不采用任何干预措施,Western blot检测大鼠十二指肠闭锁小带蛋白(ZO-1)、闭合蛋白(Occludin)表达。结果:与空白对照组比较,模型组大鼠十二指肠ZO-1、Occludin表达显著降低,十二指肠紧密连接缝隙模糊、增宽且绒毛排列不齐甚至存在断裂;与模型组比较,电针组大鼠十二指肠ZO-1、Occludin蛋白水平显著增高,十二指肠上皮细胞间紧密连接较清晰,绒毛排列整齐。结论:功能性消化不良发病与十二指肠黏膜屏障相关,电针可能通过上调十二指肠ZO-1、Occludin表达修复肠黏膜机械屏障而发挥治疗FD作用。Objective:To investigate the effect of duodenal mucosal mechanical barrier and the mechanism of Electroacupuncture(EA)in the treatment of FD.Methods:30 SD rats were randomly divided into control group,model group and EA group,each group has 10 rats.The model group and EA group were modeled by multi-factor intervention method for 20 days,and the control group wasn t dealt.After modeled,the EA group received treatment for 14 days,while the rest group did not adopt any intervention measures.After treatment,the expressions of ZO-1 and Occludin were detected by Western blot.At the same time,the ultra microstructure was detected by Electron Microscopy.Results:Compared with the control group,the ZO-1 and Occludin expression in the model group were significantly reduced,and the tight junction gaps were blurred,widened,and the villi were not arranged in an orderly manner or even fractured.Compared with the model group,Occludin and ZO-1 were significantly increased in the EA group,and the tight junctions were relatively clear,with orderly villi arrangement.Conclusion:The pathogenesis of FD has much relationship with duodenal mucosal barrier.EA may play an important role in the treatment of FD by upregulating the expression of ZO-1 and Occludin in duodenum and repairing the mechanical barrier of intestinal mucosa.
