摘 要:对腊肠树果实中的重要潜在外来有害生物粗鳞蜡螟(Trachylepidia fructicassiella Ragonot)的幼虫和成虫进行了DNA条形码研究.通过提取粗鳞蜡螟幼虫和成虫样品的基因组DNA,设计ITS1、ITS2、16S rRNA、CYT-B、COⅠ、COⅡ基因引物,PCR扩增并测序,结合Genbank中相关数据,借助生物信息学分析软件对比分析目的基因序列相似性,构建NJ系统进化树,分析遗传进化关系,获取该虫DNA条形码。结果表明,从粗鳞蜡螟幼虫和成虫的快速准确鉴定,获得了其幼虫和成虫多个基因的DNA序列,共计16条。其中,14条为新序列,分析表明这些序列可作为粗鳞蜡螟新的DNA条形码序列,为该虫的识别鉴定和幼虫的早期防治提供基础数据。The DNA barcodes of the larvae and adults of Trachylepidia fructicassiella, an important potential alien pest for China, were studied to facilitate its rapid and accurate identification. We extracted the genomic DNA of larvae and adult samples and amplified the ITS1, 16 S rRNA, CYT-B, COⅠ, and COⅡ genes by PCR. The amplified products were sequenced and aligned to GenBank sequences. The unknown larvae and adult samples were identified as Trachylepidia fructicassiella through COI sequence alignment, and 14 out of the 16 sequences amplified were new to the database.Sequence divergences were calculated and a neighbor-joining tree of distances was created. The DNA barcodes of this species were obtained by evolutionary relationships analysis. The results showed that these new sequences of ITS1, ITS2,16 S rRNA, CYT-B, and COII could be used as DNA barcodes of Trachylepidia fructicassiella for identification and early control of the larvae.
