南方红豆杉提取物对人前列腺癌细胞增殖及凋亡的影响

摘　　要：目的探讨南方红豆杉提取物对人前列腺癌细胞增殖及凋亡的影响。方法采用MTT法检测不同浓度(0、0.1、0.2、0.4、0.8、1.5、3、6μg/μL)南方红豆杉提取物对人前列腺癌细胞株DU145和PC-3增殖的抑制作用,分别计算IC50。使用南方红豆杉提取物0、1.5μg/μL分别处理PC-3和DU145细胞,并于第1~7天收集细胞进行存活细胞计数。检测不同浓度(0、1、2、3μg/μL)南方红豆杉提取物对PC-3细胞凋亡及周期的影响。结果不同浓度南方红豆杉提取物对DU145和PC-3细胞均有抑制作用(P<0.05),且随着药物浓度的增加,其抑制作用逐渐增强。PC-3细胞的IC50为2.39μg/μL,DU145细胞的IC50为3.84μg/μL。南方红豆杉提取物1.5μg/μL对PC-3和DU145细胞均有抑制作用,并且随着作用时间的延长,抑制作用逐渐增强。与南方红豆杉提取物0μg/μL相比,南方红豆杉提取物2、3μg/μL处理PC-3细胞处于早期凋亡与晚期凋亡的数量和G1期细胞比例逐渐增加(P<0.05)。结论南方红豆杉提取物能抑制人前列腺癌细胞增殖,使肿瘤细胞周期出现G1期阻滞,促进其凋亡。Objective To explore the effect of Taxus chinensis var.mairei on the proliferation and apoptosis of human prostate cancer cells.Methods MTT method was used to detect the inhibitory effects of Taxus chinensis var.mairei in different concentrations(0,0.1,0.2,0.4,0.8,1.5,3 and 6μg/μL)on the proliferation of human prostate cancer DU145 and PC-3 cells,and the half inhibitory concentrations(IC50)were calculated,respectively.PC-3 and DU145 cells were treated with Taxus chinensis var.mairei in the concentrations of 0 and 1.5μg/μL and collected from the 1st to 7th day for viable cell counts.The effects of different concentrations(0,1,2 and 3μg/μL)of Taxus chinensis var.mairei on PC-3 cell apoptosis and cycle were tested.Results Taxus chinensis var.mairei had inhibitory effects on DU145 and PC-3 cells in different concentrations(P<0.05),and the inhibition increased gradually with the increase of dose.The IC50 values of PC-3 and DU145 cells were 2.39μg/μL and 3.84μg/μL,respectively.Taxus chinensis var.mairei had inhibitory effects on PC-3 and DU145 cells in the concentration of 1.5μg/μL,and the inhibitory effect gradually increased with the prolongation of the intervention time.Compared with Taxus chinensis var.mairei 0μg/μL treatment,the apoptosis number of cells in the early and late stages and the proportion of cells at G1 phase gradually increased after PC-3 cells were treated with Taxus chinensis var.mairei in the concentrations of 2 and 3μg/μL(P<0.05).Conclusion Taxus chinensis var.mairei can inhibit the proliferation of human prostate cancer cells,block the tumor cell cycle at G1 phase,and promote the apoptosis of human prostate cancer cells.
