摘 要:目的研究双蓣调脂汤对高脂血症模型大鼠的降血脂作用及其作用机制。方法40只SD雄性大鼠随机分为5组,即空白组、模型组、双蓣高、低剂量组、辛伐他汀组。空白组饲以普通饲料,其余大鼠采用高脂饲料喂养4周建立高脂血症模型。造模成功后,双蓣高、低剂组分别给予双蓣调脂汤15.6 g/kg、7.8 g/kg灌胃,辛伐他汀组给予辛伐他汀4 mg/kg灌胃,用药治疗8周。以生化法测定各组大鼠血清总胆固醇(TC)和甘油三酯(TG),HE染色观察肝组织形态学的变化,采用RT-PCR法检测肝组织中固醇调节元件结合蛋白2(SREBP-2),固醇调节元件结合蛋白1c(SREBP-1c)和肝X受体α(LXR-α)mRNA的含量;采用Western blot法和免疫组化法检测SREBP-2、LXR-α蛋白的表达。结果与模型组相比,双蓣调脂汤能显著降低TC和TG水平(P<0.01),改善肝细胞脂肪变性;同时能够显著降低SREBP-2,SREBP-1c和LXR-α(P<0.05)的表达。结论结论双蓣调脂汤可以有效降低高脂血症大鼠血脂水平和改善肝脏脂肪变性,同时能够通过抑制SREBP-2、SREBP-1c和LXR-α的表达而抑制TC和TG的合成,这可能是双蓣调脂汤治疗高脂血症的机制之一。Objective To clarify the hypolipidemic effects of Shuangyu Tiaozhi decoction on hyperlipidemic rat model and its mechanism.Methods Forty SD rats were randomly divided into the basic control group,the model group,Shuangyu Tiaozhi high and low dose group,Simvastatin group.The basic control group was fed with common foods and the remaining rats were fed with the high-fat diet to built hyperlipidemic model for four weeks.After the successful modeling,Shuangyu Tiaozhi high(15.6 g/kg)and low(7.8 g/kg)dose group,Simvastatin group(4 mg/kg)were applied by intragastric administration for 8 weeks.The ser-um content of cholesterol(TC)of and triglyceride(TG)were detected by automated chemistry analyzer.Morphological changes of liver were observed using HE stain.The mRNA expression levels of sterol regulatory element binding protein-2(SREBP2),sterol regulatory element binding protein-1c(SREBP-1c)and liver X receptor-α(LXR-α)were detected using RT-PCR.The protein expression levels of SREBP-2 and LXR-αwere detected using Western blotting and immunohistochemistry.Results Compared with model group,Shuangyu Tiaozhi decoction obviously reduced TC and TG(P<0.01),significantly im-proved hepatic steatosis,remarkably decreased the expression levels of SREBP-2,SREBP-1c and LXR-α(P<0.05).Con-clusion Shuangyu Tiaozhi decoction effectively regulates the lipid metablism of hyperlipidemic rats and liver function.Shuangyu Tiaozhi decoction can inhibit the synthesis of TC and TG by down-regulating the expression of SREBP-2,SREBP-1c and LXR-α,which may be the mechanisms of treating hyperlipidemia.
