摘 要:【目的】探究不同正番茄斑萎病毒(Tomato spotted wilt orthotospovirus,TSWV)分离物中NSs与其地理来源和寄主二者之间的相关性,阐明云南不同地区8个烟草样品TSWV NSs适应性进化情况。【方法】利用RT-PCR、克隆、转化和测序技术,从云南不同地区已经确定感染TSWV的8份烟草样品中获得NSs氨基酸序列。采用DNAMAN 8.0和MEGA 7.0软件,对云南不同地区8个烟草样品与GenBank公布的烟草、番茄和辣椒上31个TSWV NSs氨基酸序列进行比对分析。通过The Selecton Server软件,对云南不同地区8个烟草样品TSWV NSs氨基酸序列进行选择压力分析。【结果】云南不同地区8个烟草样品与同样来自中国云南烟草、番茄和辣椒上TSWV NSs氨基酸序列在系统发育树中处于同一支,亲缘关系最近;与来自韩国的番茄和辣椒上TSWV NSs氨基酸序列位于同一大支,亲缘关系较近;而与来自欧洲保加利亚烟草上TSWV NSs氨基酸序列处于不同分支,亲缘关系最远。在云南不同地区8个烟草样品TSWV NSs氨基酸序列中未检测到正选择位点。【结论】不同TSWV分离物中NSs与其地理来源有明显的相关性,但跟寄主的相关性不大。云南不同地区8个烟草样品TSWV NSs稳定的保持其增强病毒症状、抑制沉默等功能。[Purpose]To explore the correlation between NSs in different isolates of Tomato spotted wilt orthotospovirus(TSWV)and their geographical origin and host,and to clarify the adaptive evolution of TWSV NSs in eight tobacco samples from different regions of Yunnan.[Method]The NSs amino acid sequence was obtained from eight tobacco samples infected with TSWV in different regions of Yunnan Province by RT-PCR,cloning,transformation and sequencing techniques.Eight tobacco samples from different regions of Yunnan Province were compared with the amino acid sequences of 31 TSWV NSs on tobacco,tomatoes and peppers published by GenBank using DNAMAN 8.0 and MEGA 7.0 software.Selective pressure analysis was performed on the amino acid sequences of eight tobacco samples TSWV NSs in different regions of Yunnan by the Selecton Server software.[Result]The amino acid sequences of eight tobacco samples from different regions of Yunnan and TSWV NSs from Yunnan tobacco,tomatoes and peppers in China were in the same branch in the phylogenetic tree,and the genetic relationship was the closest.It is located in the same branch as the amino acid sequence of TSWV NSs on tomatoes and peppers from South Korea,and is closely related;however,it is in a different branch from the amino acid sequence of TSWV NSs from European Bulgarian tobacco,and has the closest relationship.No positive selection sites were detected in the amino acid sequences of eight tobacco samples TSWV NSs in different regions of Yunnan Province.[Conclusion]There was a significant correlation between NSs and their geographical origin in different TSWV isolates,but the correlation with the host was not significant.Eight tobacco samples TSWV NSs in different regions of Yunnan maintained their functions of enhancing viral symptoms and inhibiting silencing.
