摘 要:目的:观察鼠曲草提取液对3,4-苯并芘大鼠肺内注射诱发肺癌的抑制作用,并初步探讨其机制。方法:将120只SD大鼠随机分为对照组和高、中、低浓度组,每组30只。经胸壁肺内注射3,4-苯并芘2 mg/次,每周1次,共4次,诱发肺部肿瘤。对照组正常饮水,高、中、低剂量组分别以高、中、低浓度鼠曲草提取液代水喂食,于半年后处死全部大鼠,取其肺组织标本,观察各组成瘤率差异。采用原位末端转移酶标记检测肿瘤细胞凋亡情况;采用逆转录-聚合酶链反应法检测大鼠肺癌组织survivin表达情况。结果:高、中、低浓度组大鼠肺癌成瘤率分别为36.67%(11/30)、43.33%(13/30)、55.17%(16/29),而对照组大鼠肺癌成瘤率为72.41%(21/29)。原位末端转移酶标记检测发现鼠曲草提取液可显著促进肺癌细胞的凋亡,且与浓度相关。大鼠肺癌组织survivin表达显示,鼠曲草提取液可显著降低大鼠肺癌组织中survivin表达。结论:鼠曲草提取液对大鼠肺癌形成有明显抑制作用。Objective:To study the inhibitory effect of Gnaphalium affine extracts on the formation of lung cancer induced by intrapulmonary injection of 3,4-benzopyrene in Rats,and to explore its mechanism.Methods:120 SD rats were randomly divided into four groups,with 30 rats each.Lung tumor was induced by intrapulmonary injection of 3,4-benzopyrene,2 mgeachtime,once a week for 4 times.The control group was given normal drinking water.High,medium and low doses groups were fed with high,medium and low concentrations of Gnaphalium affine extracts instead of water feeding.After half a year,all rats were killed and lung tissue samples were taken to observe the difference of tumor formation rates of each group.The apoptosis of tumor cells was detected by TUNEL assay,the expression of survivin in rat lung cancer tissues was detected by RT-PCR.Results:The tumor formation rates of lung cancer in high,medium and low concentration groups were 36.67%(11/30)、43.33%(13/30)and 55.17%(16/29)respectively,while the tumor formation rate of lung cancer in the control group was 72.41%(21/29).TUNEL detection showed that the extracts of Gnaphalium affine could significantly promote the apoptosis of lung cancer cells,and it was related to concentration.The expression of survivin in lung cancer tissue of rats showed that the extract of Gnaphalium affine could significantly reduce the expression of survivin in lung cancer tissue of rats.Conclusion:The Gnaphalium affine extracts has a significant inhibitory effect on lung cancer formation in rats.
