摘 要:目的:分析川芎茶调散的活性成分,预测其治疗偏头痛的作用靶点及信号通路,解析其效应机制。方法:通过TCMSP数据库收集川芎茶调散活性成分并进行ADME筛选,收集活性成分潜在靶点;通过GeneCards数据库和DisGeNET数据库收集偏头痛相关的疾病靶点,并与药物活性成分靶点取交集得到共同靶点,进行PPI分析;使用Cytoscape构建可视化"药物-靶点-疾病"网络并进行分析,通过R语言对共同靶点进行GO富集与KEGG富集分析。结果:通过筛选获得141个药物与疾病的共同靶点,发现川芎茶调散治疗偏头痛的关键靶点为APP、CHRM2、CXCL8、CXCL2、CXCL10等;主要改变的信号通路为PI3K-AKT信号通路、HIF-1信号通路,内分泌抵抗信号通路等;氧化应激反应、类固醇激素反应、血管收缩调节、低氧反应、雌二醇反应和神经递质代谢可能是川芎茶调散治疗偏头痛的主要生物学调控环节。结论:槲皮素、木犀草素、山柰酚等是川芎茶调散中重要活性成分,APP,CHRM2、CXCL8等基因受到了药物调控,川芎茶调散可能通过PI3K-AKT信号通路,HIF-1信号通路,内分泌抵抗信号通路调控了偏头痛时的炎症反应、血管收缩、内分泌-神经递质代谢等生物过程。OBJECTIVE To analyze the active components of Chuanxiong Chatiao Powder, predict its therapeutic target and signal pathway for migraine, and unveil its effect mechanism.METHODS The active components of Chuanxiong tea were collected by TCMSP database and screened by ADME to collect the potential targets of active components;the disease targets related to migraine were collected by GeneCards database and DisGeNET database, and intersected with the active drug targets to obtain common targets for PPI analysis;the visual ‘drug-target-disease’ network was constructed and analyzed using Cytoscape, and GO enrichment and KEGG enrichment analysis of common targets were performed by R language.RESULTS total of 141 common targets were obtained. The key targets for the treatment of migraine were APP, CHRM2, CXCL8, CXCL2, CXCL10, etc. The major signaling pathways were PI3 K-AKT signaling pathway, HIF-1 signaling pathway, endocrine resistance signaling pathway, etc. Oxidative stress response, steroid hormone response, vasoconstriction regulation, hypoxia response, estradiol response and neurotransmitter metabolism may be the main biological regulatory links in the treatment of migraine with Chuanxiong Chatiao Powder.CONCLUSION Quercetin, luteolin and kaempferol etc. are important active ingredients of Chuanxiong Chatiao Powder. Genes such as APP, CHRM2, CXCL8 etc. might be regulated by these active components. Chuanxiong tea powder may regulate inflammatory response, vasoconstriction, endocrine-neurotransmitter metabolism and other biological processes in migraine through PI3 K-AKT signaling pathway, HIF-1 signaling pathway and endocrine resistance signaling pathway.
