摘 要:目的:探讨榄香烯乳对小鼠全脑缺血再灌注(ischemia/reperfusion,I/R)损伤的影响。方法:采用结扎小鼠双侧颈总动脉10 min再灌注24 h制作全脑I/R模型,小鼠分为假手术组、模型组、榄香烯乳组。给药组于术前和术后30 min两次腹腔注射榄香烯乳。评价术后24 h小鼠神经症状和神经功能变化;采用苏木素-伊红染色(HE)观察神经元的病理学改变和神经元密度;TUNEL染色法观察皮层神经元凋亡;ELISA试剂盒检测血清IL-1β、IL-6和TNF-α水平。结果:榄香烯乳能改善术后24 h神经症状评分(P<0.05)和神经功能评分(P<0.001),皮层神经元密度提高了31.1%;榄香烯乳明显减少皮层神经元凋亡,较模型组减少了110.8%,同时抑制炎症因子表达(P<0.001)。结论:榄香烯乳可通过抑制凋亡和炎症水平而减轻小鼠全脑缺血再灌注损伤。Objective:To explore the effect of elemene on mice with global cerebral ischemia-reperfusion(I/R)injury.Methods:The mice cerebral I/R model was established by ligation of bilateral common carotid artery for 10 minutes and reperfusion for 24 hours.The mice were randomly divided into sham-operation group,model group(I/R)and therapy group(I/R+elemene).Elemene was intraperitoneally injected into mice of therapy group before and after I/R surgery.After 24 h of surgery,changes of neurological symptoms and functions in mice were evaluated.The changes of pathology and density of neurons were assayed by HE staining,apoptosis of cortical neurons was detected by TUNEL assay,and the blood levels of IL-1β,IL-6 and TNF-αwere detected by enzyme linked immunosorbent assay(ELISA).Results:After 24 h of surgery,elemene could significantly improve the neurological symptom score(P<0.05)and the neurological function score(P<0.001)of mice,increase the neurons density by 31.1%,decrease the apoptosis of cortical neurons by 110.8%compared with the model group and inhibit the expression of inflammatory factors(P<0.001).Conclusion:Elemene could improve the global cerebral ischemia-reperfusion injury of mice through inhibiting apoptosis and decreasing the level of inflammation factors.
