摘 要:目的:建立RP-HPLC法测定人血清中泊沙康唑的浓度。方法:采用乙腈沉淀蛋白法对血样进行预处理,色谱条件为Agilent ZORBAX SB-C18分析柱(4.6 mm×250 mm,5μm),流动相为乙腈∶水(60∶40),检测波长为262 nm,流速1.0m L·min^-1,柱温25℃,内标为阿普唑仑。结果:泊沙康唑的线性范围为0.125~50.000μg·m L^-1(r=0.9996),方法回收率为99.80%,日间和日内RSD均<4%,绝对回收率为94.58%。对照品血清在室温放置24 h和–20℃冰箱保存10 d的稳定性RSD均<3%。测定患者血清中泊沙康唑药物浓度为1.18μg·m L-1。结论:本方法快速、准确、可靠,适用于泊沙康唑血药浓度的检测。Objective:To establish a method for determining the concentration of posaconazole in human serum by reversed-phase high performance liquid chromatography(RP-HPLC).Methods:The blood samples were pretreated by acetonitrile precipitation.The chromatographic column was Agilent ZORBAX SB-C18 analytical column(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile and water(60∶40).The detection wavelength was 262 nm.The flow rate was 1.0 mL·min^-1.The column temperature was set as 25℃.And the internal standard was alprazolam.Results:The linear ranges of posaconazole were 0.125–50.000μg·mL^-1(r=0.9996),and the method recovery rates were 99.80%.The intra-day and inter-day relative standard deviations(RSD)were all less than 4%.The absolute recovery rates were 94.58%.The stability RSDs of control serum placed at room temperature for 24 h and stored in the refrigerator at–20℃for 10 day were all less than 3%.The serum concentration of posaconazole of the patient determined by this method was 1.18μg·mL-1.Conclusion:The method was fast,accurate and reliable,which was suitable for determining the blood concentration of posaconazole.
