摘  要:[目的]本文旨在克隆茄子(Solanum melongena L.)促分裂原活化蛋白激酶(MAPK)基因SmNTF 3,阐明其在盐和干旱胁迫处理下的表达特性。[方法]以茄子‘苏崎1号’为试验材料,克隆SmNTF 3的全长序列,对其基因结构和序列同源性进行分析,通过烟草叶片瞬时表达系统进行SmNTF3蛋白的亚细胞定位,采用实时荧光定量PCR检测SmNTF 3基因在不同组织及盐和干旱胁迫下的表达情况。[结果]SmNTF 3基因开放阅读框长度为1119 bp,编码1个含372个氨基酸残基的蛋白。生物信息学分析表明,该蛋白具有保守的S_TKc结构域,富含α-螺旋和无规则卷曲。系统发育分析表明,SmNTF3属于MAPK家族C组成员,其氨基酸序列具有高度保守性,与番茄和马铃薯亲缘关系最近。亚细胞定位发现,SmNTF3蛋白在烟草叶片的细胞核和细胞质膜上表达。SmNTF 3基因具有组织特异性,在茎中表达量最高。盐和干旱胁迫能够显著诱导SmNTF 3基因的上调表达。[结论]从茄子中克隆了1个MAPK家族C组成员SmNTF 3,该基因受盐和干旱胁迫诱导表达,可能参与茄子对盐和干旱胁迫的响应过程。[Objectives]The paper aimed to clone mitogen-activated protein kinase(MAPK)gene SmNTF 3 in eggplant and detect its expression patterns characteristics under salt and drought stresses.[Methods]The full-length of SmNTF 3 was cloned from eggplant‘Suqi 1’.The gene structure and sequence homology were analyzed by bioinformatics methods.Subcellular localization of SmNTF3 protein was performed with the tobacco leaf transient expression system.The expression patterns of SmNTF 3 gene in different organs and under salt and drought stresses were analyzed using real-time quantitative PCR technique.[Results]The open reading frame of the eggplant SmNTF 3 gene was 1119 bp in length,and encoded a protein of 372 amino acid residues with a conserved S_TKc domain and the richα-helices and irregular curls.Phylogenetic analysis showed that SmNTF3 belonged to a member of MAPK family,and the amino acid sequence of SmNTF3 was highly conserved and had the closest relationship with tomato and potato.Subcellular localization analysis revealed that SmNTF3 protein was located on the nucleus and plasma membrane of tobacco leaves.The expression level of SmNTF 3 was in different tissue-specific and high in stem,and increased under the salt and drought treatments.[Conclusions]In this study,SmNTF 3,a member of MAPK family C,was cloned from eggplant,and was induced by salt and drought stresses.The results indicated that it may be involved in the response of eggplant to salt and drought stresses.
