摘 要:绵羊的产羔性状研究对于提升绵羊繁育力至关重要,彭波半细毛羊作为西藏第一个国家级绵羊新品种,对其产双羔的机制研究能够有力推动藏系绵羊的人工繁育工作.采用基于TMT标记的定量蛋白质组学方法,对发情期产双羔彭波半细毛羊卵巢的特异表达蛋白和调控机制进行系统研究.共检测到6446种彭波半细毛羊卵巢蛋白,其中双羔组有116种蛋白相比单羔组出现了上调表达,171种蛋白出现了下调表达.通过对差异表达蛋白的功能分析,发现产双羔绵羊卵巢中蛋白的表达合成及运输等生物过程显著增强,同时细胞的增殖、运动及信号转导等生理活动也更加活跃.另一方面,产双羔绵羊卵巢中的脂肪酸降解、脂类代谢、氨基酸等小分子化合物的降解和代谢明显下降.此外,产双羔绵羊卵巢细胞中的糖酵解活动有所增强,而三羧酸循环有所减弱.上述结果表明产双羔彭波半细毛羊卵巢内的蛋白表达调控机制能满足更强的卵泡发育和排卵活动需求,从而促进卵泡的发育及成熟卵子的排出;一些有机小分子催化酶出现了明显的特异表达,作为其底物的小分子有机物可以作为产双羔的潜在标志物进行更深入的研究;结果可促进对彭波半细毛羊产双羔性状及藏系绵羊繁育力的深入研究.A study of litter size is essential to improve the breeding ability of sheep.As the first new national breed of sheep in Tibet,China,studies on the mechanism of the twin-lamb trait of Pengbo semi-fine wool sheep will strongly facilitate artificial breeding in Tibetan sheep.In this study,a quantitative proteomics method based on TMT labeling was used to study the differentially expressed proteins in the estrus ovary of twin lambproducing Pengbo semi-fine wool sheep.A total of 6446 proteins were found in the ovary of Pengbo semi-fine wool sheep,of which 116 proteins were upregulated and 117 proteins were downregulated in the twin lambing group(compared with the single lambing group).These differentially expressed proteins were further analyzed by GO and KEGG pathway analysis;the results showed that the following terms:protein biosynthesis and transport,cell proliferation,cell motivation,cell migration,and signal transduction pathways were enhanced,while the degradation and metabolism of fatty acids,lipids,amino acids,and other small molecular compounds were downregulated.In addition,the glycolytic activity in the ovary of twin lambs-producing sheep increased,while the activity of the tricarboxylic acid cycle was attenuated.These results demonstrate that the regulation mechanisms of protein expression in the ovary of sheep producing twin lambs could meet the stronger needs of follicular development and ovulation activity,thus promoting the development of follicles and the discharge of mature eggs.Moreover,the specific expression of some organic small molecule catalytic enzymes indicates that these molecules can be further developed into potential markers for the twin-lambing trait.Taken together,these results are not only beneficial to the research of litter size of Pengbo semi-fine wool sheep,but also to the fecundity research in Tibetan sheep and could help to further promote the development of animal husbandry in Tibet.
