摘 要:目的:探索脑肠肽在胃窦、结肠、下丘脑组织与血清中的表达以及差异性,为脑肠肽参与食欲中枢-摄食行为调控提供实验依据.方法:于2018年7月选择SPF级SD大鼠10只,适应性喂养3 d,麻醉后取胃窦、结肠、下丘脑组织及血清,ELISA法检测组织和血清中的LP、NPY、CCK-8的蛋白表达,比较在各组织中、组织与血清中的表达.结果:比较脑肠肽在胃窦、结肠及下丘脑各组织中的表达,LP蛋白在下丘脑中表达最高,三组比较差异有统计学意义(P<0.01);NPY蛋白在结肠中表达最高,三组比较差异有统计学意义(P<0.01);CCK-8蛋白在下丘脑中表达最高,三组比较差异有统计学意义(P<0.01).比较脑肠肽在各组织与血清中的表达,LP蛋白在血清中表达最高,在血清中的表达均显著高于在各组织中,差异均有统计学意义(P<0.01);NPY蛋白表达在结肠组织最高,在结肠组织中的表达显著高于血清中,差异有统计学意义(P<0.01);CCK-8蛋白表达在下丘脑组织最高,在下丘脑组织中表达显著高于血清中,差异有统计学意义(P<0.01).结论:LP、NPY、CCK-8在大鼠胃窦、结肠、下丘脑组织与血清中均有表达,共同参与食欲中枢与摄食行为的调控.表达量存在一定的差异性,与其分泌部位和途径有关联性,表达量高的脑肠肽可能发挥主导作用.Objective:To explore the expression and difference of brain-gut peptide in gastric antrum,colon,hypothalamus and serum,to provide experimental basis for brain-gut peptide to participate in the regulation of appetite center-feeding behavior.Method:In July 2018,10 SPF SD rats were selected and given an adaptive feeding regimen for 3 d.After anesthesia,gastric antrum,colonic hypothalamus tissues and serum were taken.The protein concentration of LP,NPY,CCK-8 in tissues and serum were detected by ELISA,the expression of LP,NPY and CCK-8 in tissues and serum were compared.Result:Compared the concentration expression of NCP in gastric antrum,colon and hypothalamus,LP protein concentration was the highest in the hypothalamus,the difference in three groups was statistically significant(P<0.01);the highest expression of NPY protein was found in the colon,the difference was statistically significant(P<0.01);the expression of CCK-8 protein was highest in the hypothalamus,the difference among three groups was statistically significant(P<0.01).By comparing the expression of NCP in each tissue and serum,LP protein expression was the highest in serum,the expression in serum were significantly higher than those in each tissue,the differences were statistically significant(P<0.01);the expression level of NPY protein was the highest in colon tissue,the expression level was significantly higher in colon tissue than in serum,with statistically significant difference(P<0.01);the expression of CCK-8 protein was the highest in the hypothalamus,the expression was significantly higher in the hypothalamus than in the serum,the difference was statistically significant(P<0.01).Conclusion:LP,NPY and CCK-8 are expressed in gastric antrum,colon,hypothalamus and serum,it involves in the regulation of appetite center-feeding behavior.There is a certain difference in the expression level,which is correlated with the secretion site and pathway,the high expression level of brain-gut peptide may play a leading role.
