摘 要:目的:研究尖叶假龙胆在各剂量下对酒精性肝损伤(ALD)模型大鼠的保护作用机制。方法:60只健康雄性Wistar大鼠随机分为6组,通过梯度酒精灌胃法制造大鼠ALD模型,给予对应组动物阳性对照溶液或不同剂量尖叶假龙胆提取液灌胃,8周后测定ALD模型大鼠血清中的血清脂联素(ADPN)、肿瘤坏死因子-ɑ(TNF-ɑ)、游离脂肪酸(NEFA)的含量以及肝组织中乙醛脱氢酶(ALDH)、乙醇脱氢酶(ADH)、丙二醛(MDA)的浓度水平,比较各组之间的成分的差异,并通过HE染色切片观察肝组织病理变化。结果:与空白组相比,尖叶假龙胆给药后,ALD模型大鼠病理反应明显减轻,血清中ADPN浓度升高、TNF-α、NEFA的浓度降低,肝组织中ALDH、ADH浓度升高、MDA水平降低。结论:尖叶假龙胆对ALD具有保护作用,其机制可能与增加脂质代谢,加快肝脏分解代谢酒精,减少酒精对肝脏造成的氧化损伤有关。Objective:To study the protective mechanism of Gentiana acuminata on alcoholic liver damage(ALD)rats.Methods:60 healthy male Wistar rats were randomly divided into 6 groups.The ALD model was made by gradient alcohol gavage.The interventions groups were given positive control solution or different doses of Gentiana acuminata extract correspondingly.After 8 weeks,the serum levels of adiponectin(ADPN),tumor necrosis factor-α(TNF-α)and non-esterified fatty acid(NEFA),as well as the concentrations of ALDH,ADH and MDA in liver tissues were detected and compared among the groups.The pathological changes of liver tissues were observed by HE staining.Results:Compared to those in the blank group,the pathological response of ALD model rats was significantly reduced,the serum level of ADPN was increased,and the serum levels of TNF-αand NEFA were decreased,the concentrations of ALDH and ADH were heightened and the concentration of MDA was lowered after the intervention of Gentianella acuta.Conclusion:Gentiana acuminata has a protective effect on ALD,and its mechanism may be related to the increase of lipid metabolism,the acceleration of liver metabolism of alcohol,and the reduction of oxidative damage caused by alcohol.
