摘 要:目的:壳聚糖作为人体可吸收的多糖类多聚物,与其他制作缓释药物的材料相比具有更好的生物学优势,本实验将其用于制备可注射性微球,并对其粒径、形态进行检测和观察,为其在生物医学领域的应用提供依据。方法:(1)以乙酸处理的壳聚糖作为分散相,以正辛烷、Span、戊二醛配制的溶液作为连续相和固化相,利用微流控技术制备得到油包水结构的壳聚糖微球;(2)通过尝试得到最佳收集方法;(3)进一步对其形态结构、大小尺寸、流动性、稳定性、可注射性进行研究。结果:(1)选用1.6 mm T型管生成的微球体积较小;(2)当连续相和固化相的配比均为9.0 mL正辛烷、0.8 mL Span和0.2 mL戊二醛时,既可以节省材料又可以使微球更加稳定、流动性好且具有较好的可注射性;(3)用梯度换水的方法处理制得的微球,保证其流动性,稳定性及可注射性。结论:采用微流控技术成功制备出可注射性壳聚糖微球,微球形态均匀,不粘连,粒径均一,制备方法可重复性好。Objective:As a polysaccharide polymer that can be absorbed by the human body,chitosan has better biological advantages than other materials for preparing slow-release drugs.In this experiment,chitosan was used to prepare injectable microspheres.The particle size and morphology were mainly detected,which may provide a basis for its application in the biomedical field.Methods:(1)Acetic acid-treated chitosan was used as the dispersed phase,and a solution prepared by span,n-octane,and glutaraldehyde was used as the continuous and solid phases.The water-in-oil chitosan microspheres were prepared by microfluidic technology.(2)The best collection effect was obtained by gradient water exchange method.(3)The morphology,size,fluidity,stability,and injectability were studied.Results:(1)The results showed that the use of a 1.6T T-tube could produce microspheres with smaller size.(2)When the ratio of the continuous phase and the solid phase were both 9.0 mL n-octane,0.8 mL Span and 0.2 mL pentane in the case of dialdehyde,it could not only save materials,but also make the microspheres more stable with good fluidity and better injectability.(3)To treat the prepared microspheres with a gradient water exchange method could ensure the fluidity,stability and injectability of the microspheres.Conclusion:The injectable chitosan microspheres were successfully prepared by microfluidics,the microspheres were uniform in shape,non-adhesive,uniform in particle size,and the preparation method was reproducible.
