摘 要:目的:优化对坐叶总三萜的提取工艺,测定对其含量。方法:采用正交优化,紫外分光光度法,根据L9(34)正交表,以料液比、溶剂浓度、提取时间级提取次数为四个因素进行实验,得出其最佳提取工艺,并进行其不同产地的对坐叶药材的总三萜含量测定。结果:最佳提取工艺为料液比为1∶30,溶剂浓度为80%乙醇溶液,提取次数为1次,提取时间为60 min。在最佳提取工艺条件下,不同产地对坐叶总三萜平均含量分别为贵定地区10.96 mg/ɡ、铜仁地区9.30 mg/ɡ、黔东南地区7.73 mg/ɡ、安顺地区11.46 mg/ɡ。结论:本实验方法简单、可操作性强、重复性高,可为对坐叶药材质量标准提供实验依据。In order to optimize the extraction process of total triterpenes from Hedyotis uncinella Hook.et Arn., the content of triterpenes was determined. In this experiment, orthogonal optimization and ULTRAVIOLET spectrophotometry were used. According to L9(34) orthogonal table, the optimum extraction technology was found by taking the ratio of material to liquid, solvent concentration and extraction time as four factors, and the total triterpene content of the herbs from different places of origin was determined. The results showed that the optimal extraction process was the solid-liquid ratio of 1∶30, the solvent concentration of 80 % ethanol solution, the extraction times of once, and the extraction time of 60 min. Sitting on the best extraction technology conditions, different regions average total triterpenoid content respectively set in 10.96 mg/ɡ, 9.30 mg/ɡ,’s tongren region, province area 7.73 mg/ɡ, 11.46 mg/ɡ, anshun area. Therefore, the experimental method is simple, maneuverable and repeatable, which can provide experimental basis for the quality standard of Hedyotis uncinella Hook.et Arn..
