摘 要:目的分析1,2-二羟基蒽醌降低金黄色葡萄球菌与白念珠菌混合生物膜的作用及其机制。方法收集2018年临床分离鉴定的2株耐甲氧西林金黄色葡萄球菌(MRSA)以及2株甲氧西林敏感金黄色葡萄球菌(MSSA);利用分光光度计法检测1,2-二羟基蒽醌作用后金黄色葡萄球菌与白念珠菌DAY185混合菌群生长能力变化,并进一步采用96孔板结晶紫染色法测定1,2-二羟基蒽醌作用前后金黄色葡萄球菌与白念珠菌DAY185单独生长时以及混合生物膜形成能力差异。结果1,2-二羟基蒽醌可以明显降低金黄色葡萄球菌与白念珠菌DAY185混合菌群的生长能力;静态生物膜实验显示1,2-二羟基蒽醌可以明显降低金黄色葡萄球菌与白念珠菌DAY185混合生物膜的形成能力,其作用主要是通过降低白念珠菌DAY185的生物膜形成能力。结论1,2-二羟基蒽醌可以降低金黄色葡萄球菌与白念珠菌混合生物膜形成的能力,其机制主要通过降低白念珠菌菌丝形成的相关基因ALS3和RBT1的表达。Objective To investigate the inhibitory effect of 1,2-dihydroxyanthraquinone on the formation of polymicrobial biofilms by Staphylococcus aureus and Candida albicans.Methods Two methicillin-resistant S.aureus(MRSA)strains and two methicillin-sensitive S.aureus(MSSA)strains were collected from patients in 2018.Microbial growth of mixed S.aureus and C.albicans DAY185 strains was compared before and after addition of 1,2-dihydroxyindole using a spectrophotometer.The biofilm formation ability was measured for S.aureus or C.albicans DAY185 alone and for the mixture of S.aureus and C.albicans on 96-well plates using crystal violet staining method with or without addition of 1,2-dihydroxyanthraquinone.Results The growth of S.aureus and C.albicans strains in the mixture was reduced significantly after addition of 1,2-dihydroxyanthraquinone.Moreover,1,2-dihydroxyanthraquinone could reduce the formation of polymicrobial biofilms by S.aureus and C.albicans DAY185,primarily by reducing the biofilm formation ability of C.albicans DAY185.Conclusions The compound 1,2-dihydroxyanthraquinone can reduce the formation of polymicrobial biofilms by the mixture of S.aureus and C.albicans,primarily by inhibiting the biofilm formation ability of C.albicans.This effect may be due to the reduced expression of ALS3 and RBT1 genes,which are involved in the formation of C.albicans hypha.
