摘 要:目的探究清肾颗粒对慢性肾衰竭(chronic renal failure, CRF)大鼠肾组织细胞自噬及凋亡的影响和作用机制。方法 30只大鼠随机分为对照组、CRF组和CRF+清肾颗粒组,每组10只。通过腺嘌呤构建CRF模型,CRF+清肾颗粒组大鼠给予清肾颗粒灌胃,对照组、CRF组给予等量生理盐水灌胃。检测3组肾组织损伤情况、肾功能指标,通过TUNEL检测肾组织细胞凋亡情况,通过Western blot检测肾组织细胞自噬水平,观察对核因子-κB(NF-κB)信号通路的影响。结果与对照组比较,CRF组尿素(BUN)、肌酐(Scr)、肾组织细胞凋亡率和NF-κB、NF-κB抑制因子α(IκBα)蛋白水平显著升高,而自噬标志蛋白Beclin1和LC3水平显著降低(P<0.05)。CRF+清肾颗粒组BUN、Scr水平、肾组织细胞凋亡指数和NF-κB、IκBα蛋白水平低于CRF组,Beclin1和LC3水平高于CRF组(P<0.05,P<0.01)。结论清肾颗粒可以通过抑制NF-κB通路诱导肾组织细胞的自噬,从而抑制凋亡,改善CRF肾功能。Objective To investigate the effect and mechanism of Qingshen Granules on autophagy and apoptosis in chronic renal failure(CRF)rats.Methods Thirty rats were randomly divided into control group,CRF group and CRF+Qingshen granule group,with 10 rats in each group.CRF model was established by adenine.Rats in CRF+Qingshen granule group were given Qingshen granule by gavage,while the control group and CRF group were given the same amount of normal saline.Renal tissue injury and renal function indexes were detected.Apoptosis was detected by TUNEL and autophagy level was detected by Western blot.The effect on nuclear factor-κB(NF-κB)signaling pathway was observed.Results Compared with the control group,the levels of blood urea nitrogen(BUN),creatinine(Scr),apoptosis rate of renal tissue,NF-κB and NF-κB inhibitory factorα(IκBα)protein levels in CRF group were significantly increased,while the levels of Beclin1 and LC3 were significantly decreased(P<0.05).The levels of BUN,Scr,apoptosis index of renal tissue,NF-κB and IκBαprotein were lower in CRF+Qingshen granule group than in CRF group,whereas Beclin1 and LC3 levels were higher than those in CRF group(P<0.05,P<0.01).Conclusion Qingshen Granules can induce autophagy in renal tissue cells by inhibiting the NF-κB pathway,thereby inhibiting apoptosis and alleviating renal function in the CRF rats.
