摘  要:目的通过检测铅中毒大鼠股骨的骨量、生物力学指标以及病理骨形态计量等指标,阐明铅暴露对大鼠骨质疏松的影响,并进一步研究染料木素作为防治铅暴露致大鼠骨质疏松药物的可行性,为铅中毒的效应研究以及防治提供实验依据。方法大鼠醋酸铅饮水染毒建立模型,染料木素通过灌胃的方式进行给药干预。通过原子吸收分光光度计检测各组SD大鼠股骨铅含量,运用X线骨密度仪检测大鼠骨密度,运用INSTRON 1195电子拉伸实验机检测大鼠股骨生物力学指标,进而探讨铅暴露对大鼠骨质疏松的影响,以及染料木素的保护作用。结果铅暴露大鼠股骨干重、灰重、骨钙及骨密度(bone mineral density,BMD)与对照组相比显著降低,给予染料木素干预后,其股骨干重、灰重以及BMD明显升高(P<0.05);铅暴露大鼠股骨的最大载荷(maximum loading,MaxL)、最大桡度(maximum strain,MaxS)、弹性载荷(flexibility loading,FleL)和弹性桡度(flexibility strain,FleS)等指标均有明显下降,用染料木素干预后其生物力学指标均有明显改善,差异有统计学意义(P<0.05)。结论慢性铅暴露可以引起大鼠股骨的骨质疏松,染料木素对铅致骨质疏松有一定的保护作用。Objective To elucidate the effect of lead exposure on osteoporosis in rats through detecting femoral bone,biomechanical indexes and pathological bone histomorphometry indexes of rats with lead poisoning,to further study the feasibility of genistein as prevention and treatment medicine for osteoporosis induced by lead exposure in rats,and to provide laboratory evidence for effect study on lead poisoning and its prevention and treatment.Methods Rats were exposed to lead acetate in drinking water to establish lead poisoning models,and genistein was administered by gavage intervention.Atomic absorption spectrophotometer was used to determine lead content of femur in SD rats of each group.The bone mineral density(BMD)of rats was detected by X-ray absorptiometry.The INSTRON1195 electronic tensile test machine was employed for detecting biomechanical indexes of femoral bone of the rats,and then the effect of lead exposure on osteoporosis in the rats and the protective effect of genistein were explored.Results The values of femur dry weight,ash weight,bone calcium and BMD of lead-exposed rats decreased significantly as compared with the control group,but the values of femur dry weight and ash weight and BMD increased significantly(P<0.05)after administration of genistein.Lead-exposed rats’indexes like femoral maximum loading(MaxL),maximum strain(MaxS),flexibility loading(FleL)and flexibility strain(FleS)significantly decreased,but the biomechanical indexes were improved significantly after the intervention with genistein,showing statistically significant differences(all P<0.05).Conclusions Chronic lead exposure can cause femur osteoporosis in rats.Genistein has some protective effects on lead-induced osteoporosis.
