补肾活血祛风方对阿霉素大鼠外周血淋巴细胞多耐药基因MDR1及P糖蛋白170的影响及意义  被引量:1

摘  要:目的:观察补肾活血祛风方对阿霉素肾病大鼠外周血淋巴细胞多耐药基因(multidrug resistance gene 1,MDR1)及P糖蛋白170(P-glycoprotein 170,P-gp170)的影响。方法:设空白对照(A组),阿霉素(adriamycin,ADR)肾病大鼠随机均分为模型组(B组)、泼尼松组(C组)、中药组(D组)、中药+泼尼松组(E组),每组20只。除空白组,其余分别予蒸馏水、予醋酸泼尼松片溶液、补肾活血祛风汤剂、泼尼松和汤剂灌胃。4周末时用PCR法检测各组外周淋巴细胞MDR1mRNA,8周末加测P-gp170。结果:(1)各干预组4周末MDR1mRNA差异无统计学意义,但均高于空白组(P<0.05);8周末,泼尼松组、中药+泼尼松组MDR1mRNA高于模型组(P<0.05),中药组MDR1mRNA明显低于泼尼松组(P<0.01);泼尼松组8周末MDR1mRNA高于4周末(P<0.01),其余组前后差异无统计学意义。(2)8周末,泼尼松组P-gp170明显高于中药组、中药加泼尼松组、模型组(P<0.01)。(3)外周血淋巴细胞MDR1mRNA和P-gp170呈正相关(r=0.466,P=0.001)。结论:(1)阿霉素肾病大鼠外周血淋巴细胞MDR1mRNA与P-gp170高于正常大鼠,且具有线性相关,醋酸泼尼松能使二者进一步升高。(2)补肾活血祛风方不引起阿霉素大鼠外周血淋巴细胞MDR1升高,能降低P-gp170水平,但这能否视为其减轻耐药,提高醋酸泼尼松效果的机制仍待进一步研究。Objective:To investigate the effect of decoction that could tonify kidney and invigorate the blood circulation,drive the wind,on the Multidrug resistance gene 1(MDR1)and P glycoprotein 170(P-gp170)of peripheral-blood lymphocyte in rats with adriamyc in nephropathy.Methods:SD rats were set up normal group(A),SD rats within adriamyc nephropathy were divided into model group(B),prednisone group(C),decoction group(D),prednisone and decoction(E),20 rats pergroup.At the weekend of 4 and 8,selecting ten rats randomly from each group to observe the MDR1mRNA in peripheral lymphocytes by PCR.Besides,measuring the P-gp170 with flow cytometry at the 8th weekend.Results:(1)At the 4th weekend no Statistical difference appearanced among the four intervention groups about the MDR1mRNA,but higher than group A.At the 8th weekend,MDR1mRNA of peripheral-blood lymphocyte in prednisone group(group C)and prednisone-decoction group(groupE)were higher than model group(group B)(P<0.05),MDR1mRNA in decoction group(groupD)obviously lower than prednisone group(group C).Meanwhile,MDR1mRNA in prednisone group(group C)at 8th weekend were obviously higher than the 4th weekend(P<0.01).(2)P glycoprotein 170(P-gp170)of peripheral-blood lymphocyte inprednisone group(group C)were obviously higher than that in group B,group D and group E(P<0.01).(3)There existed positive correlation between MDR1mRNA and the P-gp170 in peripheral blood lymphocytes(r=0.466,P=0.001).Conclusion:(1)MDR1mRNA and the P-gp170 in peripheral blood lymphocytes in rats with adriamyc in nephropathy were higher than the normals,there existence linear correlation between them.Inprednisone could rise both of them,even more visible with the time going.(2)Decoction that could tonify kidney,invigorate the blood circulation and drive the wind,would not bring increasion of MDR1 in peripheral blood lymphocytes rats with adriamycin nephropathy.As well,it could reduce the expression of P-gp170.So,whether this may be the mechanism of the decoction to enhance the prednisone effection still nee
