摘  要:通过克隆测序获得广东虫草(Tolypocladium guangdongense)PRO1(TgPRO1)序列并对其相关序列进行分析,进一步利用定量PCR研究子实体发育不同时期以及菌丝体在不同光照(红光、绿光、蓝光、白光、黑暗)、温度处理下TgPRO1表达情况。结果表明:TgPRO1长度为2135 bp,含有一个内含子,其CDS全长为2010 bp,编码669个氨基酸。蛋白同源性分析表明TgPRO1蛋白序列与已报道的PRO1蛋白序列高度相似。TgPRO1启动子区包含茉莉酸甲酯、光、植物激素响应元件等多种顺式作用元件。与菌丝期比较,TgPRO1在原基期表达量最高;不同光照条件下培养时,黑暗条件TgPRO1表达量较高;与23℃处理3 h比较,17℃处理3 h时TgPRO1表达量显著上调。The transcription factor gene PRO1 from Tolypocladium guangdongense(TgPRO1)was cloned and analyzed for its expression at different fruiting body development periods,under different light treatments(red light,green light,blue light,white light,and darkness)and different temperature treatments by qRT-PCR.The results showed that TgPRO1 was 2135 bp with one intron.Its complete coding sequence(CDS)was 2010 bp,encoding 669 amino acids.Protein homology analysis showed that TgPRO1 was highly homologous to other PRO1 proteins reported in previous studies.The promoter region of TgPRO1 contained cis-acting elements,e.g.MeJA response elements,light response elements and plant hormone response elements.The expression level of TgPRO1 was the highest at the primordium stage.In terms of light condition,the expression level of TgPRO1 was higher in darkness than other light conditions.The expression level of TgPRO1 was significantly up-regulated after treatment at 17℃for 3 h compared with that of the normal cultivation temperature at 23℃for 3 h.
