摘  要:目的:评价益肾通脉方改善脑缺血再灌注大鼠神经功能方面的作用。方法:将雄性SD大鼠随机分为假手术组、模型组、益肾通脉低剂量组和益肾通脉高剂量组;采用线栓法制备大脑中动脉梗死缺血再灌注模型(缺血时间为1h),再灌注24 h后对造模成功大鼠分别给药,假手术组、模型组给予等量蒸馏水。观察各组大鼠神经功能缺损变化及血清MDA、SOD、HIF-1α、ET-1、eNOS、NO水平。结果:益肾通脉低剂量组大鼠的神经功能缺损评分在第2、3、4、5天显著高于假手术组(P<0.05),但与模型组无统计学差异(P>0.05)。益肾通脉高剂量组大鼠的神经功能缺损评分在前两天时明显高于假手术组,但低于模型组。与假手术组相比,SOD的含量在模型组和益肾通脉低剂量组中显著下降,而HIF-1α却显著增加。结论:益肾通脉方可改善脑缺血再灌注大鼠神经功能,升高血清SOD水平,降低血清HIF-1α和e NOS含量,提示其脑保护机制可能与抗氧化、减轻炎症反应有关。Objective To evaluate the effect of Yishentongmai Recipe on improving neurological function in rats with cerebral ischemia-reperfusion.Methods male SD rats were randomly divided into sham operation group,model group,Yishentongmai low-dose group and Yishentongmai high-dose group;The middle cerebral artery infarction(MCAO)ischemia-reperfusion model was prepared by thread embolism method(ischemia time was 1 h).After 24 h reperfusion,the rats were given medicine respectively,and the sham operation group and model group were given equal amount of distilled water.The changes of nerve function defect and serum levels of MDA,SOD,HIF-1α,ET-1,e NOS and NO were observed.Results the scores of neurological deficit in the Yishentongmai low-dose group were significantly higher than those in the sham operation group on the 2 nd,3 rd,4 th and 5 th days(P<0.05),but there was no significant difference between the two groups(P>0.05).In the first two days,the neurological deficit score of the high-dose Yishentongmai group was significantly higher than that of the sham operation group,but lower than that of the model group.Compared with the sham operation group,the content of SOD decreased significantly in the model group and Yishentongmai low-dose group,but HIF-1αincreased significantly.Conclusion Yishentongmai decoction can improve the neurological function of rats with cerebral ischemia-reperfusion,increase the level of SOD in serum,and decrease the content of HIF-1αand e NOS in serum.It suggests that the brain protection mechanism of Yishentongmai decoction may be related to antioxidation and reducing inflammatory reaction.
