摘 要:目的:使用大孔吸附树脂分离纯化西藏猫乳总黄酮,并对纯化后的总黄酮进行体外抗氧化活性研究。方法:通过静态吸附实验考察ADS-F8、D101、AB-8等8种大孔吸附树脂对西藏猫乳总黄酮的纯化效果,并研究样液质量浓度、p H值、上样体积对树脂吸附率的影响以及洗脱液浓度、体积对洗脱率的影响。采用DPPH法、ABTS法和FRAP法,评价纯化后西藏猫乳总黄酮的抗氧化活性。结果:采用ADS-F8型树脂对西藏猫乳进行分离纯化,最佳工艺条件为:上样浓度0.6 mg/mL,p H为3.0,体积23 BV,洗脱剂为60%的乙醇,洗脱体积8 BV。所得西藏猫乳总黄酮纯度由17.18%提高至24.67%,精制倍数为1.44。抗氧化实验结果表明,纯化后西藏猫乳总黄酮对ABTS自由基的清除率可以达到99.15%,DPPH自由基清除率也可达到67.84%,体现出良好的自由基清除能力,并对Fe^3+有较好的还原力,FRAP值最高为939.8μmol/L。结论:ADS-F8型树脂可对西藏猫乳总黄酮进行分离纯化,提高总黄酮纯度,纯化后的总黄酮具有较强的抗氧化作用。Ojective Separation and purification of total flavonoids from Rhamnella gilgitica Mansf.et Melch.by macroporous adsorption resin,and the antioxidant activity of the total flavonoids was studied in vitro.Methods Purification of total flavonoids from Rhamnella gilgitica Mansf.et Melch.by eight macroporous adsorptive resins such as ADS-F8,D101 and AB-8,was investigated by static adsorption experiments,and the effect of sample liquid mass concentration,p H value and sample volume on resin adsorption rate was investigated,as well as the effect of eluate concentration and volume on elution rate.The antioxidant activity of purified total flavonoids from Rhamnella gilgitica Mansf.et Melch.was evaluated by DPPH,ABTS and FRAP methods.Results Separation and purification of Rhamnella gilgitica Mansf.et Melch.by ADS-F8 Resin,the optimum technological parameters as follows:the concentration of the sample 0.6 mg/m L,p H 3.0,the volume 23 BV,the eluting regent 60%ethanol,and the volume of elution 8 BV.The total flavone content of Rhamnella gilgitica Mansf.et Melch.increased from 17.18%to 24.67%,and the refining multiple was 1.44.The results of antioxidant test showed that the scavenging rate of ABTS and DPPH free radicals of total flavonoids from Rhamnella gilgitica Mansf.et Melch.could reach 99.15%and 67.84%respectively,which showed good free radical scavenging ability and good reducing power to Fe^3+.The highest FRAP value was 939.8μmo L/L.Conclusion ADS-F8 resin can be used to separate and purify the total flavonoids from Rhamnella gilgitica Mansf.and improve the purity of total flavonoids.The purified flavonoids have strong antioxidant effect.
