摘　　要：目的建立高效液相色谱波长切换法同时测定葆肾合剂中绿原酸、咖啡酸、芒果苷、薏苡素、鞣花酸及毛蕊异黄酮葡萄糖苷的含量。方法色谱柱Waters XBridge C18(250 mm×4.6 mm,5 μm);流动相乙腈(A)-0.2%磷酸水溶液(B),梯度洗脱;流速1.0 mL·min-1;柱温30℃;进样体积10 μL;检测波长为326 nm(绿原酸、咖啡酸、芒果苷、薏苡素)和254 nm(鞣花酸、毛蕊异黄酮葡萄糖苷)。结果该条件下所测6种成分的色谱峰分离度较好,其他成分对测定无干扰。绿原酸、咖啡酸、芒果苷、薏苡素、鞣花酸和毛蕊异黄酮葡萄糖苷分别在3.26~74.34 μg·mL-1(r=0.9999)、1.72~39.20 μg·mL-1(r=0.9999)、1.61~36.85 μg·mL-1(r=0.9998)、2.17~36.34 μg·mL-1(r=0.9999)、1.65~37.65 μg·mL-1(r=0.9999)、3.28~74.96 μg·mL-1(r=0.9999)与峰面积线性关系良好,平均加样回收率均大于96.0%,RSD均小于3.0%。结论建立的方法准确可靠、操作简便、重复性好,可用于葆肾合剂的质量控制。Objective To determine the content of chlorogenic acid,caffeic acid,mangiferin,coixol,ellagic acid and pistil isoflavone glucoside in Baoshen mixture by HPLC wavelength switching method.Methods The Waters XBridge C18 column (250 mm×4.6 mm,5 μm) was used.The mobile phase consisted of acetonitrile (A)-0.2% phosphoric acid aqueous solution (B) with gradient elution.The volume flow was 1.0 mL·min-1,the column temperature was 30℃,the sample volume was 10 μL,and the detection wavelength was 326 nm (for chlorogenic acid,caffeic acid,mangiferin,and coixol) and 254 nm (for ellagic acid and pistil isoflavone glucoside).Results The 6 components were separated well without interference.The linearity of chlorogenic acid,caffeic acid,mangiferin,coixol,ellagic acid and pistil isoflavone glucoside was 3.26~74.34 μg·mL-1 (r=0.9999),1.72~39.20 μg·mL-1 (r=0.9999),1.61~36.85 μg·mL-1 (r=0.9998),2.17~36.34 μg·mL-1 (r=0.9999),1.65~37.65 μg·mL-1 (r=0.9999) and 3.28~74.96 μg·mL-1 (r=0.9999),respectively.The average recoveries were all higher than 96.0% with RSDs less than 3.0%.Conclusion The method is accurate,reliable,simple and reproducible,which can be used for the quality control of Baoshen mixture.
