小柴胡汤抗丙型肝炎病毒机制的实验研究

摘　　要：目的探讨小柴胡汤抗丙型肝炎病毒(HCV)的作用机制。方法选取ICR小鼠40只为实验对象,随机分为给药组和对照组,每组20只。给药组给予15 g/kg(ig)试药,对照组给予等量0.9%氯化钠溶液。采用不同剂量生药和不同采集时间的药物血清,处理PBK-HCV重组体转染的SMMC7721肝癌细胞,通过RT-PCR半定量法测定细胞内重组体mRNA瞬时表达情况。结果在动物实验中,小柴胡汤组T淋巴细胞增殖比对照组显著增高,T细胞亚群无显著变化。小柴胡汤对SMMC7721细胞生长的半数抑制剂量为75 mg/mL,但不能抑制重组体mRNA表达。结论小柴胡汤的抗HCV作用不是通过抑制mRNA的转录,而主要是通过调节机体免疫功能发挥作用。Objective To explore the mechanism of Xiaochaihu Decoction against hepatitis C virus(HCV).Methods Forty ICR mice were selected as experimental objects,and they were randomly divided into administration group and control group,with 20 mice in each group.The administration group was given 15 g/kg(ig)test drug,and the control group was given the same amount of 0.9%sodium chloride solution.Different doses of crude drugs and different collection time of drug serum were used to treat SMMC7721 liver cancer cells transfected with PBK-HCV recombinants,and the transient expression of recombinant mRNA in the cells was determined by RT-PCR semi-quantitative method.Results In animal experiments,the proliferation of T lymphocytes in the Xiao Chaihu Decoction group was significantly higher than that in the control group,and there was no significant change in T cell subsets.The half inhibitory dose of Xiaochaihu Decoction on the growth of SMMC7721 cells was 75 mg/mL,but it could not inhibit the expression of recombinant mRNA.Conclusion The anti-HCV effect of Xiaochaihu Decoction is not through inhibiting mRNA transcription,but mainly through regulating the body's immune function.
