摘 要:目的观察天麻素对单侧迷路破坏(Unilateral labyrinthectomy,UL)后大鼠前庭内侧核(Medial vestibular nucleus,MVN)组胺受体H1、H2、H3亚型表达的影响,探讨天麻素对前庭功能损伤大鼠的干预作用及机制。方法将99只健康SD大鼠随机分为空白对照组(n=9)、假手术组(n=18)、造模组(n=72)。造模组鼓室注射氯仿破坏左侧迷路,假手术组则注射等量生理盐水。模型评价后,将造模组大鼠随机等量分为4组:模型对照组、天麻素高、中、低剂量组。天麻素注射液肌注日1次,共3天。采用RQ-PCR法、免疫组化法及Western-Blot法观察各组大鼠MVN中组胺受体H1、H2、H3 mRNA及蛋白表达水平。结果与空白对照组(1.003±0.085、1.002±0.072、1.005±0.118;0.282±0.026、0.250±0.050、0.248±0.016)比较,模型对照组(1.315±0.078、1.418±0.09、1.467±0.083;0.408±0.056、0.453±0.031、0.403±0.057)大鼠MVN中组胺受体H1、H2、H3 mRNA及蛋白表达水平上调(P<0.05);与模型对照组比较,天麻素高剂量组(3.392±0.197、3.553±0.189、3.657±0.077;0.571±0.034、0.572±0.078、0.600±0.061)大鼠MVN中组胺受体H1、H2、H3 mRNA及蛋白表达明显上调(P<0.05),均有统计学差异。结论天麻素上调前庭功能障碍大鼠MVN中组胺受体H1、H2、H3 mRNA及蛋白表达可能是其促进大鼠前庭代偿的重要机制之一。Objective To investigate effects of gastrodin on the expression of H1,H2 and H3 subtypes of histamine receptor in the medial vestibular nucleus(MVN)in rats after unilateral labyrinthine(UL)damage and its role in vestibular compensation mechanisms.Methods Healthy SD rats(n=99)were randomly divided into a normal control group(n=9),a sham operation group(n=18)and a study group(n=72).In the study group,chloroform was injected into the left tympanum to destroy the labyrinth,while the same amount of normal saline was injected in the sham operation group.After modeling confirmation,rats in the study group were randomly selected to receive high,medium or low dose of intramuscularly injected gastrodin(once a day for 3 days)or no gastrodin(control).RQ-PCR,immunohistochemistry and Western-Blot were used to evaluate expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN.Results Compared with the normal control group,expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN in modeling animals was up-regulated(P<0.05),especially in those treated with high dose of gastrodin,which was higher than in those with no gastrodin treatment(P<0.05).Conclusion Gastrodin may play an important role in vestibular compensation by up regulating expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN in the presence of vestibular dysfunction.
