山核桃仁鞣质的HPLC指纹图谱研究

摘  要:建立山核桃仁鞣质的HPLC指纹图谱。以没食子酸为参照物,采用Welchrom C18色谱柱(150 mm×4.6 mm,5μm);流动相:0.1%磷酸水溶液(A),甲醇-0.1%磷酸水溶液(V:V=9:1)(B),进行梯度洗脱;流速:0.8 mL·min^-1;检测波长:220 nm。通过“中药色谱指纹图谱相识度评价系统(2004A版)”评价相似性,进行聚类分析。建立了该药材的HPLC指纹图谱,标定24个共有峰,方法学考察结果均符合要求,不同产地的药材分为四类。该方法稳定可靠,可用于山核桃仁鞣质的质量评价。An HPLC fingerprint of tannin of walnut was established using gallic acid as reference,Welchrom C18(4.6 mm×150 mm,5μm)chromatographic column,mobile phase as 0.1%phosphoric acid solution(A)and methanol-0.1%phosphoric acid solution(9:1)(B),gradient elution,flow rate as 0.8 mL·min^-1;detection wavelength as 220 nm.By means of Chinese Medicine Chromatographic Fingerprint Acquaintance Evaluation System(2004 A Edition),its similarity was evaluated,and its cluster analysis was conducted.HPLC fingerprints of medicinal materials from 10 producing areas were established,24 common peaks were calibrated,and the results of methodology investigation were all in accordance with the requirements.Medicinal materials from 10 different origin were divided into four categories.The established fingerprint method was stable and reliable, can be used to evaluate the quality of tannin.
