摘  要:目的观察异叶青兰总黄酮(DHBF)对高血压模型大鼠肾素血管紧张素醛固酮系统(RASS)的作用,探讨DHBF降压及抑制心肌肥厚的机制。方法两肾一夹(2K1C)法建立高血压大鼠模型,术后第6周除假手术组外,随机分为4组:模型组(Model);卡托普利组;异叶青兰黄酮高剂量组(DHBF-H)、低剂量组(DHBF-L)。灌胃给药6周后,POWERLAB系统测定血流动力学指标,进行左心室病理学检查,放免法测定血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)的水平,RT-PCR、Westernblot法测定心脏组织中血管紧张素受体1(AT1)、血管紧张素受体2(AT2)mRNA相对表达量及蛋白表达。结果给药6周后,与模型组相比,血流动力学结果显示,DHBF-H组能降低颈动脉收缩压(SBP)、降低左室收缩压(LVSP)、左室舒张末期压(LVEDP)、校正后室内压最大上升速率(+dp/dt max/LVSP)(P<0.05);延长舒张时间(DD)(P<0.05);DHBF各组心脏AngⅡ分别增高了3.1%和8.1%(P>0.05),DHBF-L组肾脏AngⅡ、ALD分别下降了2.4%和10.8%(P>0.05),DHBF-H组分别下降了6.4%(P<0.05)和27.5%(P<0.01),DHBF-H组ALD明显降低(P<0.01)。结论异叶青兰总黄酮能降低血压,调节高血压大鼠的血流动力学参数,抑制心肌肥厚,与其能够抑制AT1的表达和ALD的含量有关。Objective In traditional Uyghur,Tibetan and Indian medicine,Dracocephalum heterophyllum Benth is widely used for the treatment of hypertension.In a previous study the anti-hypertension effective component were confirmed as Dracocephalum heterophyllum Benth flavone(DHBF).The present study was aimed to tests the effect of DEBF on hemodynamic parameters and RAAS(Renin-Angiotensin-Aldosterone System), such as angiotensin-Ⅱ(AngⅡ),Aldosterone(ALD),Angiotensin Ⅱ type 1(AT1) receptor and Angiotensin Ⅱ type 2(AT2) receptor.Methods Two-kidney one-clip(2K1C) hypertensive model rats were divided randomly into five groups:sham,model,captopril20 mg/kg,DHBF 300 and 600 mg/kg(n=10 for each).Drugs were administered orally daily for six weeks.Systolic blood pressure(SBP) and diastolic blood pressure(DBP) were measured before treatment and at each week.The hemodynamics examination of the heart was performed.Blood,heart and kidney samples were collected after the last treatment.The protein expression of Ang Ⅱ,ALD,AT1 and AT2 receptors in these tissues were detected by western blot.Results After 6 weeks of administration,compared with model group,hemodynamic results showed that,in the DHBFH group,the carotid SBP,left ventricular systolic blood pressure(LVSP),left ventricular end-diastolic blood pressure(LVEDP),and the maximum rate of increase of corrected ventricular blood pressure(+dp/dt Max/LVSP) were reduced(P <0.01,P <0.05),while the diastolic time(DD) was prolonged(P <0.05).In the DHBF-H and DHBF-L group,the Ang Ⅱ of heart was increased by 3.1% and 8.1%,respectively(P> 0.05).In DHBF-L group,Ang Ⅱ and ALD of kidney were reduced 2.4% and 10.8% respectively(P> 0.05).In DHBF-H group,Ang Ⅱ and ALD of kidney were reduced by 6.4%(P <0.05) and 27.5%(P <0.01).Conclusion DHBF can improve the hemodynamic parameters of hypertensive rats,reduce blood pressure,and possible related to the inhibition of the expression of AT1 receptor and content of ALD.
