摘  要:目的采用1H核磁共振代谢组学技术研究丹红注射液干预治疗自发高血压(SHR)大鼠尿液代谢物的特征。方法12周龄SHR大鼠随机分为:高血压组(SHR组)、丹红注射液干预组(DH组);同龄Wister-Kyoto大鼠(WKY组)为正常对照组,每组7只。DH组腹腔注射10 mL/kg体质量丹红注射液,WKY组和SHR组腹腔注射等量的无菌生理盐水,连续给药28 d后检测各组大鼠血压、心率,收集24h尿液,1H-NMR测定尿液中代谢指纹图谱,PLS-DA进行模式识别,通过OPLSDA的相关系数寻找差异标记物,并对差异标记物进行KEGG分析。结果SHR组大鼠与WKY组大鼠相比较,尿液中乙酸、N-乙酰糖蛋白、柠檬酸盐、α-葡萄糖、胍基乙酸、组氨酸、α-酮戊二酸、苯基乙酰甘氨酸、赖氨酸、马尿酸含量降低,牛磺酸含量升高;DH组大鼠与SHR组大鼠相比较,尿液中乙酸、N-乙酰糖蛋白、柠檬酸盐、α-葡萄糖、胍基乙酸、组氨酸、α-酮戊二酸、苯基乙酰甘氨酸、赖氨酸、马尿酸含量升高,牛磺酸含量降低;这些代谢物的产生主要涉及到糖酵解糖异生、脂肪细胞因子信号转导通路、精氨酸和脯氨酸代谢、三羧酸循环等代谢通路。结论丹红注射液能调节大鼠尿液中多种代谢物的排泄,改善机体内环境,对机体产生保护效应。Objective To investigate the effect of DanHong injection on the urine metabolites of spontaneously hyper⁃tensive rats(SHR)by 1HNMR metabonomics technique.Methods Twelve-week-age SHR rats were randomly divided into two groups:SHR group and Danhong injection intervention group(DH group),and selecting Wister-Kyoto rats of the same age with normal blood pressure as control group(WKY group),with 7 rats in each group.The DH group was treated with DanHong Injection(10mL/kg),while SHR group was treated with the same amount of sterilized saline solu⁃tion for 28 days.Blood pressure and heart rate were checked,the urine was collected for 24h after medicine treatment.Metabolic profile in urine were determined by 1HNMR metabonomic technique,PLS-DA was used in pattern recogni⁃tion,OPLS-DA correlation coefficient was used to find differences of metabolites components,and KEGG analysis was done in the markers.Results Compare to the WKY group,acetate,α-Glucose,N-acetylglycoproteins,citrate,guanidinoacetic acid,histidine,α-ketoglutaric acid,lysine,phenylacetylglycine and hippuric acid were increased,while taurine was decreased in urine of the SHR group.Compare to the SHR group,acetate,α-Glucose,N-acetylgly⁃coproteins,citrate,guanidinoacetic acid,histidine,α-ketoglutaric acid,lysine,phenylacetylglycine and hippuric acid were decreased,while taurine was increased in urine of the DH group.The production of these metabolites is mainly in⁃volved in glycolysis and gluconeogenesis,adipocytokine signal transduction pathway,arginine and proline metabolism,tricarboxylic acid cycle.Conclusions The excretion of various metabolites were regulated by DanHong injection in rat urine,which improved the internal environment and produced protect effect of the body.
