摘  要:目的研究抑制自噬对昆仑雪菊水提物抗INS-1胰岛细胞脂毒性损伤的保护作用。方法采用棕榈酸(PA)诱导建立INS-1胰岛细胞脂毒性损伤模型,分别给予昆仑雪菊水提物、自噬抑制剂3-甲基腺嘌呤(3-MA)和Compound C进行干预,采用荧光倒置显微镜观察细胞的形态变化,采用CCK8法检测不同处理组细胞活力,通过AnnexinV/PI双染检测细胞的凋亡情况,Western blotting方法检测细胞自噬标志蛋白LC3-II/LC3-I表达水平。结果与PA组比较,其他组给予昆仑雪菊处理可改善PA诱导的INS-1细胞损伤,提高细胞活力,差异有统计学意义(P<0.01)。与昆仑雪菊+PA组比较,给予自噬抑制剂3-MA或Compound C处理可加重细胞损伤,降低细胞存活率,差异有统计学意义(P<0.01)。与PA组比较,昆仑雪菊水提物干预组细胞凋亡作用收到抑制,差异有统计学意义(P<0.05),预先给予自噬抑制剂3-MA、Compound C处理INS-1细胞后,昆仑雪菊对PA诱导的INS-1细胞凋亡的保护作用受到抑制,差异有统计学意义(P<0.05)。结论昆仑雪菊可通过调节自噬发挥对胰岛β细胞脂毒性损伤的保护作用。Objective To study the protective effect of water extract of Kunlun Coreopsis tinctoria(CT) on INS-1 islet cells.Methods Palmitic acid(PA) was used to induce lipid toxicity injury model on INS-1 islet cells.The water extract of Kunlun CT,3-methyladenine(3-mA) and compound C were used to intervene.The morphological changes of INS-1 cells were observed by fluorescence inverted microscope.The cell viability was detected by CCK8 method.The apoptosis of INS-1 cells was detected by annexin V/PI double staining.LC3-Ⅱ/LC3-Ⅰ protein expression was detected by Western Blotting.Results Compared with the PA group,Kunlun CT treatment in the other groups could improve INS-1 cell damage and cell viability induced by PA(P <0.01).Compared with Kunlun CT+PA group,autophagy inhibitor 3-mA or compound C could aggravate cell damage and reduce cell survival rate(P <0.01).Compared with the PA group,Kunlun CT water extract inhibited the apoptosis of INS-1 cells(P <0.05).After the INS-1 cells were treated with autophagy inhibitor 3-mA and compound C,the protective effect of Kunlun CT on apoptosis of INS-1 cells induced by PA was inhibited(P <0.05).Conclusion Kunlun Coreopsis tinctoria can protect islet β cells from lipid toxicity by regulating autophagy.
