摘  要:为研究西南桦(Betula alnoides)中木质素生物合成途径的关键酶4-香豆酸:辅酶A连接酶的调控机制,本研究采用RT-PCR技术从西南桦中克隆得到一个具有完整开放阅读框的4-香豆酸:辅酶A连接酶基因(Ba4CL, NCBI登录号为MK959316),该基因全长1 629 bp,共编码542个氨基酸。西南桦Ba4CL与光皮桦(Betula luminifera, ACJ38668.1) 4CL的蛋白序列同源性为99.63%,与白桦(Betula platyphylla, AAV65114.1)4CL同源性为99.45%;Ba4CL基因拥有4CL基因家族的保守域及具有保守序列SSGTTGLPKGV和GEICIRG,Ba4CL与桦木属植物白桦和光皮桦聚在一起。Ba4CL基因在西南桦的根、小枝、树皮和叶片中均有表达,且在小枝中表达量最高,树皮次之。本研究为培育西南桦优良用材品种提供参考。In order to study the metabolism mechanism of the key enzyme 4-coumarate:coenzyme A ligase in lignin biosynthesis pathway of Betula alnoides,used the RT-PCR technology,a complete open reading frame of Ba4 CL gene was cloned from B.alnoides for the first time.The full-length c DNA of Ba4 CL gene was 1629 bp,encoded 542 amino acids(NCBI accession number:MK959316).The protein sequence of Betula alnoides is 99.63%similar to the Betula luminifera(ACJ38668.1)and 99.45%Betula platyphylla(AAV65114.1),respectively.Sequence alignment analysis showed that amino acids had conserved amino acid blocks SSGTTGLPKGV and GEICIRG.The phylogenetic analysis revealed that Ba4 CL clustered together with B.platyphylla and B.Luminifera.Transcription pattern analysis showed that Ba4 CL expressed in different tissues and had the highest transcript profile in twig.The results provided theoretical references for the study of some important mechanisms in the formation of birch materials and cultivated the superior varieties of B.alnoides.
