摘 要:目的:探讨电针能否加强GDNF基因修饰的NSCs移植对脊髓损伤后降低细胞凋亡蛋白的启动,改善大鼠的运动功能。方法:65只成年雌性SD大鼠,通过手术造成T11节段不完全横断损伤模型,并于造模后第2天局部移植GDNF修饰的NSCs,同时给大鼠做电针治疗。通过BBB评分系统来统计大鼠造模后双下肢活动的恢复程度;在不同的时间点取材,通过免疫组织化学染色,观察相关细胞凋亡因子表达,并以此来评价该法治疗脊髓损伤的效果。结果:实验动物双后肢活动BBB得分电针干预组与单纯细胞移植组无统计学差异,各治疗组与对照组均有统计学差异。在取材的病理切片中观察到caspase-8电针干预组与单纯细胞移植组有统计学差异,各治疗组与对照组均有统计学差异。结论:电针干预GDNF基因修饰神经干细胞移植与单纯的细胞移植相比可降低局部细胞凋亡因子的表达,增加轴突数量的恢复,保护脊髓应有的功能,但在大鼠双后肢活动能力的改善方面没有显著效果。Objective:To explore whether electro acupuneture can strengthen the GDNF gene modifed NSCs transplantation to reduce the initiation of apoptosis protein after spinal cord injury and improve the motor function of rats.Methods:65 adult female SD rats were operated to causeTllsegmental incomplete transection injury model,and GDNF-modified NSCs were locally transplantedon the second day after modeling,and the electro-acupuncture was given tothe ratsat the same time.The BBB scoring system was used to calculate the degree of recovery of the lower limbs activities of the rats after modeling,samples were taken at different time points,through immunohistochemical staining,observe the expression of related apoptosis factors,and use this to evaluate the effect of this method on spinal cord injury.Results:There was no statistical difference between the electro-acupuncture intervention group and the simple cell transplantation group in BBB scores of both hindlimbs activity,but there was a statistical difference between all treatment groups and the control group.In the pathological sections taken,there was a statistical difference between the caspase-8 electro-acupuncture intervention group and the simple cell transplantation group,and also have a statistical differences between all treatment groups and the control group.Conclusion:Compared with simple cell transplantation,electro-acupuncture intervention of GDNF genemodified neural stem cell transplantation can reduce the expression of local apoptotic factors,increase the number of axons,and protect the proper function of the spinal cord.However,there was no significant effeet on improvement of the mobility of both hind limbs in rats.
