摘 要:目的探析采血后6 h内制备的高脂血浆经不同孔径微孔滤膜滤过后各项质量指标变化。方法以2020年1—5月采集规格为400 mL/袋的全血(CPDA保存液联袋)制备的血浆作为原料血浆,运用比浊法随机选取86袋乳糜指数>5的高脂血浆,利用50 mL规格的一次性无菌注射器,令相同容量的高脂血浆分别经过0.45μm、0.22μm、0.10μm孔径的滤膜过滤,过滤前后分别测定甘油三酯(TG)、血浆总蛋白(TP)、凝血因子Ⅷ(FⅧ)含量。计量资料以均数±标准差(x±s)表示,过滤前后差异比较采用t检验,P<0.05为差异有统计学意义。结果 1高脂血浆经0.45μm滤膜过滤后,TG、TP、FⅧ含量差异均无统计学意义(P>0.05);2高脂血浆经0.22μm滤膜过滤后,TG含量差异有统计学意义(P<0.05),而TP、FⅧ含量差异均无统计学意义(P>0.05);3高脂血浆经0.10μm滤膜过滤后,TG、TP含量差异均有统计学意义(P<0.05),而FⅧ含量差异无统计学意义(P>0.05)。由此可知,0.22μm和0.10μm滤膜均有效去除高脂血浆中的甘油三酯,过滤后血浆活性成分含量均达到《全血及成分血质量要求》(GB18469-2012)(TP≥50 g/L、FⅧ≥0.7 IU/mL)。综合各项数据可得,0.10μm滤膜在高效去除甘油三酯的基础上,依然能保证血浆中的活性成分含量达到国家标准,是最适滤膜,此法有效降低了血浆报废率,对满足日益增长的用血需求具有重要的意义。结论采血后6 h内制备的高脂血浆经最适微孔滤膜过滤后,血浆质量符合GB18469-2012《全血及成分血质量要求》,微孔滤膜过滤法为高脂血浆正常化提供了可行性,可指导新技术开发。Objective To explore the changes of related quality indexes of hyperlipidemic plasma(prepared within 6 hours after blood collection)after filtration by microporous membrane of different pore sizes.Methods The plasma prepared from whole blood donation(400 mL/bag with CPDA preservation solution)from January to May 2020 was used as raw plasma.86 bags of hyperlipidemic plasma with chylous index greater than 5 were randomly selected by turbidimetric method.The same volume of hyperlipidemic plasma was respectively filtered through 0.45μm,0.22μm and 0.10μm filter membranes using 50 mL disposable sterile syringe.Triglyceride(TG),total protein(TP)and blood coagulation factorⅧ(FⅧ)were measured before and after filtration.The measurement data were expressed as mean±standard deviation(x±s)test was used to compare the difference before and after filtration by the SPSS 22.0 software.The significant difference was defined as P<0.05.Results 1)There was no significant difference in the contents of TG,TP and FⅧin hyperlipidemic plasma after 0.45μm membrane filtration(P>0.05);2)After the hyperlipidemic plasma was filtered by 0.22μm membrane,significant difference was notable in the content of TG(P<0.05),but not in the contents of TP and FⅧ(P>0.05);3)After 0.10μm membrane filtration,significant difference was notable in the contents of TG and TP(P<0.05),but not in the content of FⅧ(P>0.05).Therefore,both 0.22μm and 0.10μm filter membranes can effectively remove triglycerides from hyperlipidemic plasma,and the quality indexes of plasma after filtration meet the standard in Quality Requirements for Whole Blood and Blood Components(GB18469-2012)(referred as‘the Requirement’)(TP≥50 g/L,FⅧ≥0.7 IU/mL).0.10μm membrane,which can satisfy the Requirement after high-efficiency removal of triglycerides,was an optimal filter membrane,and contributed to reduce the rate of plasma discarding.Conclusion The hyperlipidemic plasma,prepared within 6 hours after blood collection,are found to be adhering good to the Require
