摘 要:通过中药系统药理学分析平台(TCMSP)筛选香连片的主要活性成分;通过TCMSP及Swiss Target Prediction数据库查找筛选其活性成分的对应靶点.收集有关细菌性痢疾相关疾病的靶点,映射至香连片活性成分靶点基因,构建活性成分-靶点网络及共同靶点蛋白相互作用网络(PPI).将共同靶基因进行GO功能富集与KEGG通路富集分析.从香连片中筛选出42个活性成分,19个与细菌性痢疾相关疾病的作用靶点;GO功能富集筛选得到生物学过程相关条目12条、分子功能相关条目5条及细胞组分相关条目6条;KEGG通路富集筛选得到8条相关通路,涉及钙信号通路、癌症通路、跨膜受体蛋白酪氨酸激酶信号通路等.这表明香连片可能通过钙信号通路、癌症通路、跨膜受体蛋白酪氨酸激酶信号等,对细胞增殖、凋亡、炎症反应等过程产生影响从而拥有抗细菌性痢疾相关疾病功能,此外预测中表明其作用还涉及多条癌症通路,可为其预防癌症研究提供一定依据.The corresponding targets of the active ingredients of the Xianglian tablet were screened by the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP)and Swiss Target Prediction.The targets of bacterial dysentery-related diseases were collected,mapped to the target gene of the active ingredient of Xianglian tablet,so the active ingredients-target networks were constructed,and interactive protein-protein interaction network(PPI)was constructed.The common target genes were subjected to gene ontology(GO)enrichment and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analyses.42 active ingredients and 19 targets related to anti-bacterial dysentery-related diseases were screened out.12 items related to biological process,5 items related to molecular function and 6 items related to cell components were screened out by GO function enrichment;8 pathways related to cancer pathway and osteoclast differentiation signal were screened out by KEGG pathway enrichment,etc.The results show that Xianglian tablet may play a role in the treatment of bacterial dysentery-related diseases through the influence of calcium signaling pathway,pathways in cancer and other effects on cell proliferation,apoptosis,inflammatory response.In addition,multiple cancer pathways are involved in the prediction,which may provide directions for cancer prevention research.
