摘 要:为了筛选出适合火龙果种质资源遗传多样性分析和品种鉴定的SSR核心引物,以遗传背景差异较大的8份火龙果种质资源对已报道的20对火龙果SSR引物和本课题组转录组测序开发的100对EST-SSR引物进行初步筛选,筛选出引物36对。再利用18份形态差异较大、不同来源的火龙果种质资源对初筛引物进行复筛和验证,筛选出24对SSR核心引物,并通过遗传多样性分析和聚类分析验证引物的有效性。24对核心引物对18份火龙果种质资源进行遗传多样性分析,共得到117个等位基因,平均每个位点等位基因数(Na)4.88个,有效等位基因数(Ne)为2.341,期望杂合度(He)为0.516,多态性信息含量指数(PIC)平均为0.471,说明引物的多态性较高,能够有效揭示18份火龙果种质资源的遗传多样性。筛选出的24对SSR引物,带型清晰、稳定,多态性较高,可作为核心引物用于现有的火龙果种质资源鉴定和遗传多样性分析等研究。In order to screen out suitable SSR core primers which can accurately evaluate pitaya germplasm resources genetic diversity and identify pitaya cultivars,based on the 8 pitaya germplasm resources with distant genetic relationship,20 pairs of SSR primers from the articles and 100 pairs of SSR primers developed from pitaya transcriptome were initially screened out,there were 36 pairs of primers were screened out.Then 18 pitaya germplasm resources with different geographical origins and morphological characteristics were chosen to screen the initial selected primers.Finally,a total of 24 pairs of SSR core primers were identified.The genetic diversity of 18 pitaya germplasm resources was analyzed by these core markers.As a result,117 polymorphism bands were acquired.The average number of alleles(Na),effective alleles(Ne),and expected heterozygosity(He)are 4.88,2.341 and 0.516 respectively.The average PIC(Polymorphism information content)value is 0.471.The results showed that SSR primers with high polymorphism can evaluate the genetic diversity of the 18 pitaya materials effectively,the 24 core primers can produce clear amplification results with good repeatability and high polymorphism,and can be applied to pitaya germplasm resources identification and genetic diversity analysis.
