摘 要:冷胁迫是限制植物生长和分布,降低作物产量的重要环境因子.冷驯化是温带植物对抗冷胁迫的有效手段.本研究以具有耐寒能力的高山植物牛皮杜鹃(Rhododendron chrysanthum Pall.)作为研究对象,探究冷驯化前后牛皮杜鹃的基因表达和代谢通路的变化.基于转录组数据的测序结果:常温组和驯化组的总reads分别为103951和113093.将获得的unigene在KEGG、GO、KOG、Nr、Nt、Swissprot和Pfam数据库中进行比对注释,有78.67%的unigene在Nr数据库中获得了注释,比列最高.比较常温组和冷驯化组的差异表达基因为21489,其中上调和下调的基因分别为11213和10276个.GO功能分类表明差异表达基因主要富集在“DNA为模板的转录调控”“信号转导”“细胞氧化还原稳态”“蛋白质丝氨酸/苏氨酸激酶”“RNA结合”和“细胞”;KEGG路径分析显示:差异表达基因富集在132条通路上.显著性最高的生物学过程包括:光合作用、ABC载体运输、氨基酸代谢和次生代谢物质合成等.结合差异表达分析,特别地关注了脂肪酸代谢及其信号转导通路的磷脂酶基因,筛选到了10个和脂肪酸去饱和有关的基因以及12个和磷脂酶相关的基因.研究结果对揭示牛皮杜鹃的耐寒分子机制有一定的参考意义.Cold stress is an important environmental factor that restricts plant growth and distribution and reduces crop yield.Cold acclimation is an effective means for temperate plants to resist cold stress.In this study,Rhododendron chrysanthum Pall which is a subnival alpine plant possessing strong cold tolerance was used as the cold tolerant candidate to explore the changes of gene expression and metabolic pathway after cold acclimation.The results of transcriptome sequencing showed that 103951 and 113093 reads from room temperature group and cold acclimation group were obtained,respectively.For transcriptome functional annotation,the unigenes obtained were annotated in the seven public databases(KEGG,GO,KOG,Nr,Nt,Swissprot and Pfam),and 78.67%of the unigenes were annotated in the Nr database.A total of 21489 differentially expressed genes was identified,among which the up-regulated and down-regulated genes were 11213 and 10276,respectively.The most frequently assigned GO terms were“DNA as template transcriptional regulation”,“signal transduction”,“cell redox homeostasis”,“protein serine/threonine kinase”,“RNA binding”“cell part”,“binding”,“catalytic activity”and“Cell”,KEGG pathway analysis showed that differentially expressed genes were enriched in 132 pathways.Significantly highly enriched pathways included:photosynthesis,ABC carrier transport,amino acid metabolism and secondary metabolite synthesis.Combined with differential expression analysis,special attention to genes encoding fatty acid desaturase and phospholipase associated changes were paid in mRNA expression,and 10 genes related to fatty acid desaturation and 12 genes related to phospholipase were identified.The results of the research have certain reference significance for revealing the molecular mechanism of cold tolerance of Rhododendron chrysanthum Pall.
