摘 要:目的研究芫花素对大鼠产后出血合并多器官功能障碍综合征(Multiple organ dysfunction syndrome,MODS)氧化应激及凝血指标的影响。方法把大鼠平均分为对照组(NC组),产后出血合并MODS大鼠模型组(MODS组)和产后出血合并MODS大鼠模型加芫花素治疗组(YHS组)。分别观察大鼠的氧化应激指标、凝血指标和炎性因子的表达;观察大鼠肺组织的改变,比较肺组织中的TNF-α和IL-6蛋白表达。结果与NC组大鼠丙二醛(Malondialdehyde,MDA)、一氧化氮(Nitric oxide,NO)指标相比较,MODS组指标显著升高,而和NC组大鼠的超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽(Glutathione,GSH)和过氧化氢酶(Catalase enzymes,CAT)指标相比,MODS组大鼠的指标显著降低(P<0.05);芫花素干预后的YHS组大鼠的MDA指标显著低于MODS组,同时SOD、GSH和CAT指标显著高于MODS组(P<0.05)。和NC组凝血酶时间(TT)、凝血酶原时间(PT)、纤维蛋白原(Fib)和D二聚体(D-D)指标相比较,MODS组大鼠显著增高(P<0.05),而MODS组大鼠的APTT和AT指标显著低于NC组大鼠(P<0.05);和MODS组大鼠相比较,YHS组大鼠的TT和PT的时间显著缩短,并且Fib和D-D指标明显降降低(P<0.05),而YHS组大鼠的APTT和AT指标显著高于MODS组大鼠(P<0.05);和NC组的炎性因子TNF-α和IL-6的含量相比,MODS组显著增加,而YSH组大鼠的炎性因子明显得到了显著的降低(P<0.05)。和NC组相比,MODS组大鼠的肺组织损伤明显;YHS组大鼠的肺组织情况明显好转。和NC组TNF-α和IL-6蛋白相比,MODS组显著增多(P<0.05);而JHS组的蛋白表达显著降低(P<0.05)。结论大鼠产后出血合并的MODS的氧化应激损伤可由芫花素进行抑制,并且有效提高其凝血功能。Objective To study the effects of genkwanin on oxidative stress and coagulation parameters in rats with postpartum hemorrhage complicated with multiple organ dysfunction syndrome(MODS).Methods All rats were equally divided into control group(NC group),postpartum hemorrhage combined with MODS group(MODS group)and postpartum hemorrhage combined with MODS rat model plus genkwanin treatment group(YHS group).The expression of oxidative stress index,coagulation index and inflammatory factors in the three groups were observed;the pathological changes of lung tissue and the expression of TNF-αand IL-6 protein in lung tissue of three groups were compared.Results Compared with NC group,the indexes of malondialdehyde(MDA)and nitric oxide in MODS group were significantly higher than those in NC group,while superoxide dismutase(SOD),glutathione(GSH)and peroxidase(catalase enzymes)in MODS group were significantly higher than those in NC group,Compared with MODS group,the level of MDA in YHS group was significantly lower than that in MODS group(P<0.05),while SOD,GSH and catalase were significantly higher in YHS group than in MODS group(P<0.05).Compared with NC group,the time of thrombin time(TT),prothrombin time(PT),fibrinogen(FIB)and D-Dimer(D-D)in MODS group were significantly increased(P<0.05),while APTT and above indexes of MODS group were significantly lower than those of NC group(P<0.05);compared with MODS group,TT and Pt time of YHS group were significantly shortened,and FIB and D-D indexes were significantly decreased compared with NC group,the contents of TNF-αand IL-6 in MODS group were significantly higher than those in MODS group(P<0.05),while those in YHS group were significantly decreased(P<0.05).Compared with NC group,the lung tissue injury of MODS group was obvious,and that of YHS group was significantly improved.Compared with NC group,the expression of TNF-αand IL-6 in MODS group was significantly increased(P<0.05),while the protein expression of YHS group was significantly decreased(P<0.05).Conclusion Genkw
