摘 要:目的观察依达拉奉对全身麻醉下腹部手术后老年小鼠认知功能障碍的作用。方法14月龄老年小鼠随机分为5组,对照组、生理盐水组、依达拉奉低剂量组、依达拉奉中剂量组、依达拉奉高剂量组,每组5只,对照组不接受手术;其余4组分别进行以下处理:异氟烷全身麻醉下腹部手术后,腹腔分别注射生理盐水、8 mg/kg依达拉奉、16 mg/kg依达拉奉、32 mg/kg依达拉奉连续14 d。结束后分别进行新物体识别和水迷宫实验,检测各组小鼠脑内海马组织肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)和突触后致密蛋白-95(PSD-95)表达,测定乙酰胆碱酯酶活性。结果新物体识别和水迷宫实验结果显示:与对照组相比,生理盐水组老年小鼠的分辨指数显著降低,在训练第4天,生理盐水组逃避潜伏时间时间更长;与生理盐水组相比,依达拉奉组老年小鼠的逃避潜伏时间显著降低,依达拉奉32 mg/kg可显著增加老年小鼠分辨指数和在目标象限的时间百分比(P<0.05)。各组炎性因子、突触后蛋白PSD-95表达和乙酰胆碱酯酶活性检测结果显示:与对照组相比,生理盐水组海马区炎性因子TNF-α、IL-1β和IL-6表达显著上调,脑内乙酰胆碱酯酶活性显著增加,海马体内PSD-95表达水平显著下降;与生理盐水组相比,依达拉奉组海马TNF-α的表达显著下调,依达拉奉16 mg/kg和依达拉奉32 mg/kg组海马IL-1β、IL-6表达、脑内乙酰胆碱酯酶活性显著下降,海马内PSD-95的表达水平显著上调(P<0.05)。结论依达拉奉可减轻全身麻醉下腹部手术致老年小鼠的认知障碍,其作用可能与抑制神经炎症、上调突触后蛋白表达以及提高胆碱能传递有关,且在本研究剂量范围内,依达拉奉作用效果随剂量增加而增强。Objective To explore the effects of edaravone on cognitive dysfunctions induced by abdominal surgery under general anesthesia in aged mice.Methods 14-month-old mice were randomly divided into 5 groups:tcontrol group,normal saline group,edaravone low-dose group,edaravone middle-dose group,edaravone high-dose group,5 in each group,and the control group did not receive surgery;the other four groups were treated as follows:after the operation of lower abdominal hand under isoflurane general anesthesia,normal saline,8 mg/kg edaravone,16 mg/kg edaravone and 32 mg/kg edaravone were injected intraperitoneally respectively for 14 days.Novel object recognition test and Morris water maze were conducted.The expression of tumor necrosis factorα(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),and post-synaptic dense protein-95(PSD-95)in the hippocampus of mice in each group was detected,and acetylcholinesterase activity was measured.Results The results of new object recognition and water maze experiment showed that:compared with the control group,the resolution index of the old mice in the saline group decreased significantly,and the escape latency time of the saline group was longer on the 4th day of training;compared with the saline group,the escape latency time of the old mice in the edaravone group decreased significantly,and 32 mg/kg edaravone significantly increased the resolution index and the percentage of time in the target quadrant(P<0.05).Compared with the control group,the expression of TNF-α,IL-1β and IL-6 in the hippocampus of the normal saline group was significantly increased,the activity of ache in the brain was significantly increased,and the expression of PSD-95 in the hippocampus was significantly decreased;compared with the normal saline group,the expression of TNF-αin the hippocampus of the edaravone group was significantly increased In edaravone 16 mg/kg and edaravone 32 mg/kg groups,the expression of IL-1β,IL-6,AChE activity in the brain decreased significantly,and the expression of PSD-95 in t
