摘  要:目的探讨血清巯基氧化酶1(QSOX1)水平及其基因多态性与乙肝病毒性肝癌的遗传易感性的相关性。方法本研究采用病例-对照研究方法,回顾性收集德阳市人民医院收治的200例乙肝病毒性肝癌患者(HCC)及200例种族、年龄、性别匹配的对照组为研究对象。首先利用酶联免疫吸附法(ELISA)试剂盒检测两组人群血清中QSOX1的含量;利用构象差异凝胶电泳的方法将QSOX1基因启动子区的rs11589519和rs7597位点的单核苷酸多态性进行分型;利用卡方检验判断QSOX1基因型分布频率是否符合遗传平衡定律;同时分析QSOX1基因启动子区不同位点的基因多态性和等位基因型与HCC遗传易感性的相关性。分析QSOX1基因启动子区的rs11589519和rs7597位点基因多态性与HCC患者肿瘤分期的相关性。结果 HCC组血清中QSOX1的表达含量(3.48±1.82)明显高于对照组(1.91±0.78),差异有统计学意义(P <0.05),且肿瘤分期越高,血清QSOX1的含量越高。Hardy-Weinberg遗传平衡分析表明QSOX1两个基因位点的多态性均符合遗传平衡分布(r2<0.33,P> 0.05)。HCC组QSOXl位点rs11589519基因型TT频率高于对照组(38.0%vs.25.0%),rs7597基因型CT、TT频率高于对照组(48.0%vs.16.0、32.0%vs.10.0%),差异有统计学意义(P <0.05)。HCC组QSOXl基因rs11589519和rs75972位点等位基因型T分布频率均高于对照组(65.0%vs.51.0%、56.0%vs.18.0%),差异有统计学意义(P <0.05)。QSOX1基因启动子区的rs11589519位点AA型HCC患者IIIa+IIIb比率明显高于TT型患者(P <0.05)。QSOX1基因启动子区的rs7597位点TT型HCC患者Ⅲa+Ⅲb比率明显高于CC型患者,差异有统计学意义(P <0.05)。结论 QSOX1基因启动子区的rs11589519和rs7597位点多态性与HCC遗传易感性存在相关性。Objective To investigate the correlation between serum sulfhydryl oxidase 1(QSOX1)level and its genetic polymorphism and the genetic susceptibility of hepatitis B virus liver cancer.Methods In this study,200 cases of hepatitis B virus liver cancer(HCC)and 200 cases of race,age and gender matched control group were collected in People's Hospital Of Deyang City.First,the ELISA kit was used to detect the content of QSOX1 in the serum of the two groups of people;the single nucleotide polymorphisms at the rs11589519 and rs7597 positions in the promoter region of the QSOX1 gene were typed by conformational difference gel electrophoresis;Test to determine whether the frequency of QSOX1 genotype distribution conforms to the law of genetic balance;at the same time analyze the correlation between genetic polymorphisms and alleles at different sites in the promoter region of QSOX1 gene and HCC genetic susceptibility.Finally,we analyzed the correlation between the rs11589519 and rs7597 gene polymorphisms in the promoter region of QSOX1 gene and the tumor stage of HCC patients.Results The expression level of QSOX1 in serum of HCC patients was significantly higher than that in control group(3.48±1.82 vs.1.91±0.78,P<0.05),and the higher the tumor stage,the higher the content of serum QSOX1.Hardy-Weinberg genetic balance analysis showed that the polymorphisms of QSOX1 two gene loci conformed to the genetic balance distribution(r^2<0.33,P>0.05).The frequency of rs11589519 genotype TT at QSOX1 locus of HCC group was higher than that of control group(38.0%vs.25.0%),and the frequency of CT and TT genotype of rs7597 was higher than control group(48.0%vs.16.0,32.0%vs.10.0%),the difference was statistically significant(P<0.05).The frequency of allelic T distribution at rs11589519 and rs75972 locus of QSOXl gene in HCC group was higher than that in control group(65.0%vs.51.0%,56.0%vs.18.0%),and the difference was statistically significant(P<0.05).In addition,the proportion of Ⅲa+Ⅲb in AA type HCC patients at the rs11589519 locus o
