摘 要:目的优化积雪草中积雪草苷和羟基积雪草苷的微波提取工艺。方法以液料比、乙醇体积分数、微波功率和提取时间为自变量,积雪草苷和羟基积雪草苷的提取总量为响应值,在单因素实验的基础上,采用Box-Behnken中心组合实验,以4因素3水平的响应面法(RSM),优化积雪草的微波提取工艺条件。结果积雪草微波提取的最佳工艺条件为:液料比为10∶1,乙醇体积分数为44.55%,微波功率为300 W,提取时间为3.52 min,在此条件下,积雪草中积雪草苷和羟基积雪草苷的总提取量为33.03 mg·g^-1,与理论值33.70 mg·g^-1相对误差为2.01%。结论该方法具有高效、简便的优点,可用于积雪草中积雪草苷和羟基积雪草苷的提取。Objective To optimize the microwave-assisted extraction of asiaticoside and madecassoside from Centella asiatica.Methods Liquid-solid ratio,ethanol content,microwave power and extraction time were selected as the independent variables,meanwhile,the total yields of asiaticoside and madecassoside were studied as the response value.Based on single-factor test,the optimal extraction technology was established through Box-Behnken center composite design and the method of response surface methodology(RSM)with 4 factors and 3 levers.Results The optimal extraction conditions were obtained as follows:liquid-solid ratio of 10∶1,ethanol content of 44.55%,microwave power of 300 W and extraction time of 3.52 min.Under this conditions,the total yields of asiaticoside and madecassoside were 33.03 mg·g^-1,the relative error was 2.01%compared with the predicted value of 33.70 mg·g^-1.Conclusion The method has advantages of high efficiency and simplicity,which can be used for the extraction of asiaticoside and madecassoside from Centella asiatica.
