摘 要:目的探究阿司匹林联合长春瑞滨(VNR)对人乳腺癌细胞MCF-7的作用机制。方法体外培养人乳腺癌细胞MCF-7,分为空白对照组、VNR组以及阿司匹林+VNR低、高剂量组。使用MTT法检测MCF-7细胞的增殖情况,使用流式细胞仪检测MCF-7细胞的凋亡率及周期变化,使用Western Blot和RT-PCR定性、半定量检测B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl相关X蛋白(Bax)、半胱氨酸蛋白酶3(Caspase-3)、半胱氨酸蛋白酶9(Caspase-9)、磷酸化丝氨酸苏氨酸激酶(P-Akt)和丝氨酸苏氨酸激酶(Akt)的蛋白表达量。结果MTT观察结果显示,与空白对照组比较,VNR组和阿司匹林+VNR低剂量组MCF-7细胞抑制率显著升高(P<0.01);与阿司匹林+VNR低剂量组比较,阿司匹林+VNR高剂量组MCF-7细胞抑制率有一定程度升高(P<0.05)。流式细胞仪检测结果显示,与空白对照组比较,VNR组和阿司匹林+VNR低、高剂量组细胞凋亡率均显著增加,S期细胞数量比例显著增加(P<0.01)。与空白对照组比较,VNR组Bax、Caspase-3和Caspase-9蛋白表达显著升高(P<0.01),Bcl-2和P-Akt蛋白表达显著降低(P<0.01);与VNR组比较,阿司匹林+VNR低、高剂量组Bax、Caspase-3和Caspase-9蛋白表达显著升高(P<0.01),Bcl-2和P-Akt蛋白表达显著降低(P<0.01);与阿司匹林+VNR低剂量组比较,阿司匹林+VNR高剂量组Bax、Caspase-3和Caspase-9蛋白表达显著升高(P<0.01),Bcl-2和P-Akt蛋白表达显著降低(P<0.01)。结论阿司匹林联合VNR可通过下调抑凋亡蛋白Bcl-2,上调促凋亡蛋白Bax、凋亡调控因子Caspase-3和Caspase-9蛋白表达,抑制磷脂酰肌醇3-激酶/丝氨酸苏氨酸激酶(PI3K/Akt)信号通路达到抑制乳腺癌细胞增殖、促进乳腺癌细胞凋亡的效果。Objective To explore the mechanism of aspirin combined with vinorelbine(VNR)on inhibition of human breast cancer cell line MCF-7.Methods Human breast cancer MCF-7 cells were cultured in vitro and divided into blank control group,VNR group,aspirin+VNR low dose and high dose groups.MTT assay was used to detect the proliferation of MCF-7 cells.Flow cyto-metry was used to detect the apoptosis rate and periodic changes of MCF-7 cells.Western Blot and RT-PCR were used to qualitatively and semi-quantitatively detect the protein expression levels of B cell lymphoma/leukemia-2(Bcl-2),Bcl associated X protein(Bax),cysteine proteinase3(Caspase-3),cysteine proteinase 9(Caspase-9),phosphorylation serine threonine kinase(P-Akt)and serine threonine kinase(Akt).Results MTT observation showed that compared with blank control group,the growth inhibition rate of MCF-7 cell was significantly increased in VNR group and aspirin+VNR low dose group(P<0.01).Compared with aspirin+VNR low dose group,the growth inhibition rate of MCF-7 cell in aspirin+VNR high dose group was increased to some extent(P<0.05).Flow cytometry for apoptosis rate detection showed that compared with blank control group,the apoptosis was increased significantly in VNR group,aspirin+VNR low dose and high dose groups,and the proportion of cells in S phase was increased significantly(P<0.01).Compared with blank control group,the protein expression levels of Bax,Caspase-3 and Caspase-9 in VNR group were significantly increased(P<0.01),while the protein expression levels of Bcl-2 and P-Akt were significantly decreased(P<0.01).Compared with VNR group,the protein expression levels of Bax,Caspase-3 and Caspase-9 were significantly increased in aspirin+VNR low dose and high dose group(P<0.01),while the protein expression levels of Bcl-2 and P-Akt were significantly decreased(P<0.01).Compared with aspirin+VNR low dose group,the protein expression levels of Bax,Caspase-3 and Caspase-9 were significantly increased in aspirin+VNR high dose group(P<0.01)while the protein express
