摘  要:目的探讨大耳兔骨骼肌急性钝挫伤后Wnt7a表达水平的变化与损伤修复的关系。方法选择日本大耳兔40只,随机选取5只作为对照组,其余建立急性钝挫伤模型后分为7组,各组分别在钝挫伤后1d、2d、3d、4d、5d、7d、14d取材。HE染色观察钝挫伤后腓肠肌形态学变化,RT-PCR检测Wnt7a mRNA的表达水平。结果钝挫伤后1d组可观察到肌外膜完整性被破坏,出现大量的红细胞和少量的炎性细胞;钝挫伤后2d、3d组有大量的炎性细胞浸润;钝挫伤后4d组红细胞及炎性细胞减少、巨噬细胞增多、肌纤维坏死;钝挫伤后5d组出现成纤维细胞;钝挫伤后7d组出现部分新生肌细胞以及大量成纤维样细胞,并有结缔组织填充,炎性细胞浸润减轻;钝挫伤后14d组可见肌丝扭曲,肌纤维明显恢复,纤维化增加,损伤部位初步愈合。钝挫伤后自然恢复过程中,与对照组相比,钝挫伤后1d、2d组的Wnt7a mRNA水平差异无统计学意义(P>0.05);钝挫伤后3d、4d、5d、7d组Wnt7a mRNA表达水平逐渐升高,与对照组比较差异有统计学意义(P<0.05),在钝挫伤后7d时表达最高,随后开始下降。结论Wnt7a可能参与骨骼肌损伤的早期修复过程,推测损伤修复的速度很可能与Wnt7a的表达水平密切相关,Wnt7a在骨骼肌损伤修复中发挥重要作用。Objective To analyze the relationship between the expression level of Wnt7 a and the repair of acute blunt trauma in skeletal muscle of big-eared rabbits.Methods Selected forty Japanese big-eared rabbits,five were selected as control group,the rest were established the injury models and were divided into 7 groups,1 d,2 d,3 d,4 d,5 d,7 d,14 d after acute blunt trauma.Morphological changes of injured gastrocnemius were observed by HE staining.Expression level of Wnt7 a mRNA was detected by RT-PCR.Results under the light microscope,the integrity of the outer muscle membrane was damaged,a large number of red blood cells and a small number of inflammatory cells were observed 1 d after acute blunt trauma.A large number of inflammatory cell infiltration was observed 2 d and 3 d after acute blunt trauma.There were fewer red blood cells and inflammatory cells,more macrophages and muscle fiber necrosis 4 d after acute blunt trauma.Fibroblasts began to appear 5 d after acute blunt trauma.Some new muscle cells and a large number of fibroblasts appeared,which were filled with connective tissue,and the infiltration of inflammatory cells was alleviated 7 d after acute blunt trauma.Muscle filaments were distorted,muscle fibers were obviously recovered,fibrosis was increased,and the injured site was initially healed 14 d after acute blunt trauma.During the process of natural restoration after acute blunt trauma,compared with the control group,the expression level of Wnt7 a mRNA 1 d and 2 d after acute blunt trauma had no significant difference(P>0.05).The expression level of Wnt7 a mRNA increased 3 d,4 d,5 d,7 d after acute blunt trauma,was significant different(P<0.05).The expression level of Wnt7 a mRNA 7 d after acute blunt trauma was at the highest level,then began to decline.Conclusion The results suggest that Wnt7 a may be involved in the early repair process of skeletal muscle injury.It is speculated that the speed of damage repair may be closely related to the expression level of Wnt7 a,and Wnt7 a may play an important ro
