摘 要:目的分析贵州省柯萨奇病毒A组4型(coxsackievirus A4,CVA4)分离株的基因特征。方法对2009—2017年贵州省急性弛缓性麻痹(acute flaccid paralysis,AFP)病例中分离到的19株CVA4,采用反转录-聚合酶链反应(RT-PCR)方法进行VP1区扩增并对扩增产物测序,通过MEGA 7.0软件采用邻位相接法(neighbor joining method)构建遗传进化树进行基因特征分析,可靠性通过1000 Bootstrap值评估。结果19株CVA4贵州分离株之间的的核苷酸同源性为92.0%~100.0%,氨基酸同源性为96.1%~100.0%,与原型株high point株之间的核苷酸同源性为83.3%~85.7%,氨基酸同源性为95.1%~98.4%;19株CVA4均为C2基因亚型,主要聚集在Cluster1和Cluster 2分支,其中Cluster1是2009年后贵州地区持续传播的优势群体。结论2009—2017年贵州省AFP病例中CVA4贵州分离株为C2基因亚型。Objective To understand the genetic characteristics of coxsackievirus A4(CVA4)isolated in Guizhou Province of China.Methods The VP1 genes of 19 CVA4 strains isolated from acute flaccid paralysis(AFP)cases in Guizhou Province from 2009 to 2017 were amplified using reverse-transcription polymerase chain reaction and nucleotide sequencing.A phylogenetic tree was constructed by MEGA7.0 software for genetic characterization based on neighbor joining algorithm and reliability of the phylogenetic tree was determined by bootstrap analysis with 1000 pseudo replicate datasets.Results The nucleotide and the amino acid homologies among the 19 CVA4 isolates were 92.0%-100% and 96.1%-100%,respectively,and were 83.3%-85.7% and 95.1%-98.4%compared with the CVA4 prototype(High point)respectively.All the 19 strains of CVA4 were sub-genotype of C2,mainly clustered in Cluster 1 and Cluster2.Cluster 1 is a major cluster that has been persistently and extensively circulating in Guizhou since 2009.Conclusion The sub-genotype of CVA4 isolated from AFP cases in Guizhou Province of China from 2009 to 2017 was C2.
