摘 要:为确诊福建某原种猪场保育猪群临床暴发的疑似猪繁殖与呼吸综合征(PRRS),以及了解猪繁殖与呼吸综合征病毒(PRRSV)毒株流行情况,从该猪场采集2周龄~8周龄各周龄段保育猪临床疑似发病猪全血各5份,将各周龄段5份血样各取50μL混匀,并分别命名为2W、3W、4W、5W、6W、7W、8W。利用PRRSV ORF5基因特异性引物通过RT-PCR方法进行检测,并对阳性结果进行ORF5基因序列分析。RT-PCR结果显示,2周~4周的保育猪全血中未检出PRRSV,5周~8周的保育猪全血中检出PRRSV。ORF5基因序列分析显示,5W毒株的ORF5基因与JXA1、HuN4和TJ等HP-PRRSV代表毒株同源性较高,为95.4%~95.6%,而与NADC30、VR2332、QYYZ等毒株的同源性较低,为82.7%~87.4%;6W、7W、8W毒株的ORF5基因与NADC30、FJZ03、FJY04和CHsx1401等类NADC30 PRRSV代表毒株同源性较高,为88.9%~91.6%,而与JXA1、VR2332、QYYZ等毒株的同源性较低,为83.3%~85.6%。基于ORF5基因构建的遗传进化树显示,5W毒株属于HP-PRRSV,6W、7W、8W毒株属于类NADC30 PRRSV。结果表明,该猪场保育猪存在类NADC30和HP-PRRSV毒株混合感染。研究结果可为福建省防控PRRS提供理论依据。In order to confirm the suspected outbreak of PRRS in a breeding farm of Fujian province and to understand the prevalence of PRRSV strain,5 whole blood samples clinically suspected were collected from 2-8-week-old piglets from the farm.50μL blood samples were taken from 5 blood samples of each week and mixed well,and named as 2W,3W,4W,5W,6W,7W and 8W,and so on.The specific primers of PRRSV ORF5 gene were detect the samples by RT-PCR,and the positive results were analyzed by ORF5 gene sequences.RT-PCR results showed that no PRRSV was detected in the whole blood of 2W-4W conservation pigs,and no PRRSV was detected in the whole blood of 5W-8W conservation pigs.The ORF5 gene sequence analysis showed that the ORF5 gene of strain 5W had a high homology with HP-PRRSV representative strains such as JXA1,HuN4 and TJ(95.4%-95.6%),but a low homology with NADC30,VR2332 and QYYZ(82.7%-87.4%).The ORF5 gene of strains 6W,7W and 8W had high homology with NADC30 PRRSV,such as NADC30,FJZ03,FJY04 and CHsx1401(88.9%-91.6%),while the homology with strains JXA1,VR2332 and QYYZ(83.3%-85.6%).The genetic evolution tree constructed based on ORF5 gene showed that strain 5W belonged to HP-PRRSV,and strains 6W,7W and 8W belonged to NADC30 PRRSV.The results showed that there were mixed infection of NADC30 and HP-PRRSV strains in the conservation pigs.This study provided theoretical basis for the prevention and control of PRRS in Fujian province.
