摘  要:目的:建立HPLC法测定毛竹笋中尿嘧啶、尿苷、胸腺嘧啶、腺苷的含量。方法:采用HPLC法,色谱柱为Hypersil C18柱(4.6 mm×250 mm,5μm),以甲醇-水为流动相,梯度洗脱,流速0.5 m L/min,检测波长为254 nm,柱温30℃。结果:尿嘧啶、尿苷、胸腺嘧啶、腺苷分别在5.1~102.0μg/m L、5.0~100.0μg/m L、4.9~98.0μg/m L、5.1~102.0μg/m L的范围内具有良好的线性关系(R^2分别为0.9998,0.9999,0.9997,0.9998);平均加样回收率分别为98.13%,97.55%,97.90%,98.61%;RSD分别为1.15%,0.89%,1.48%,1.92%。结论:该法简单快捷,结果准确,可用于测定毛竹笋中核苷类成分的含量。Objective:To establish the method for the determination of uracil,uridine,thymine and adenosine in shoots of Phyllostachys edulis by HPLC.Methods:HPLC method was adopted.The column was Hypersil C18(4.6 mm×250 mm,5μm),methanol and water as the mobile phase with a flow rate of 0.5 m L/min,with gradient elution,the detection wavelength was 254 nm,and the column temperature was 30℃.Results:The linear range of nucleosides were 5.1~102.0μg/m L、5.0~100.0μg/m L、4.9~98.0μg/m L、5.1~102.0μg/m L(R^2 were 0.9998,0.9999,0.9997,0.9998).The average recovery were 98.13%,97.55%,97.90%,98.61%,the RSD were 1.15%,0.89%,1.48%,1.92%.Conclusion:The method is simple,rapid and accurate,it can be used for the determination of nucleosides in shoots of Phyllostachys edulis.
