摘  要:目的基于网络药理学方法,初步探讨二仙汤治疗高血压的潜在作用机制。方法通过检索中药系统药理学数据库和分析平台(TCMSP)得到二仙汤中6味中药(淫羊藿、仙茅、巴戟天、黄柏、知母、当归)的化学成分及作用靶点,借助Cytoscape 3.6.1构建二仙汤成分-靶点网络图。在TTD、基因功能富集分析平台(CTD)、Drugbank数据库中检索得到高血压疾病靶点,结合二仙汤预测靶点,进而基于蛋白质-蛋白质相互作用(PPI)分析、基因本体(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析,探究二仙汤治疗高血压的作用机制。结果本研究共得到二仙汤候选药效成分78种,预测靶点144个,高血压相关疾病靶点333个,化学成分-疾病共同靶点53个。进行PPI分析,得到6个关键作用靶点基因,分别为eNOS、TNF、TP53、AR、JUN、PTGS2。富集结果显示,其主要与信号转导、RNA聚合酶Ⅱ启动子转录正调控等生物过程有关,可能作用于神经活性配体-受体相互作用、TNF信号通路、PI3K-Akt信号通路等。结论通过网络药理学的方法,得到了二仙汤治疗高血压6个关键作用靶点基因eNOS、TNF、TP53、AR、JUN、PTGS2,初步推测通过神经活性配体-受体相互作用、TNF信号通路等发挥作用,但有待进一步实验验证。Objective To explore the mechanism of Erxian Decoction in the treatment of hypertension based on network pharmacology.Methods The chemical components and action targets of Erxian Decoction Epimedium(Rhizoma Curculiginis,Rhizoma Anemarrhenae,Cortex Phellodendri,Morinda Officinalis,Angelica)were obtained by searching the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The component-target network diagram of Erxian Decoction was constructed by Cytoscape 3.6.1.The targets of hypertension were retrieved from TTD,CTD,and drugbank databases.The targets were combined with the predicted targets of Erxian Decoction.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genes Genomes(KEGG)pathway enrichment analysis to explore the mechanism of Erxian Decoction in the treatment of hypertension based on protein-protein interaction(PPI)analysis.Results A total of 78 candidate pharmacodynamic components,144 predicted targets,333 hypertension targets,and 53 common targets of chemical components and diseases were obtained.Six key targets were obtained through PPI analysis.The enrichment results showed that PPI was mainly related to signal transduction,RNA polymeraseⅡpromoter transcription positive regulation and other biological processes,which might act on the neural active ligand-receptor interaction,TNF signal pathway,PI3K-Akt and other signal pathways.Conclusion The mechanism of Erxian Decoction in the treatment of hypertension is explored through the method of network pharmacology,and 6 key targets of eNOS,TNF,TP53,AR,Jun and PTGS2 were obtained.It is preliminarily speculated that of Erxian Decoction acts through the interaction of neuroactive ligands and receptors,TNF signaling pathway.However,further experimental verification was needed.
