摘 要:目的:采用HPLC-DAD同时测定柴胡口服液中柴胡皂苷a和柴胡皂苷d的含量。方法:采用CAPCELL PAKC18高效液相色谱柱(4.6 mm×250 mm,5μm),乙腈—水(1%三乙胺)梯度洗脱做为流动相,流速1.0 ml/min,检测波长210nm。结果:柴胡口服液中的柴胡皂苷a、柴胡皂苷d分别在1.27~126.43μg、1.08~135.45μg范围内显良好的线性关系。结论:采用HPLC-DAD法同时测定柴胡口服液中的柴胡皂苷a和柴胡皂苷d,具有操作简单、重复性好、回收率高、峰形对称等优点,可以作为柴胡口服液的含量测定方法。Objective: Simultaneous determination of saikosaponin a and saikosaponin d in Chaihu oral solution by HPLC-DAD. Methods: A CAPCELL PAK C18 high performance liquid chromatography column(4.6 mm×250 mm, 5 μm) was used, and the acetonitrile-water(1% triethylamine) gradient elution was used as the mobile phase. The flow rate was 1.0 ml/min, and the detection wavelength was 210 nm. Results: The content of saikosaponin a and saikosaponin d in Bupleurum oral liquid showed a good linear relationship in the range of 1.27~126.43 μg and 1.08~135.45 μg, respectively. Conclusion: Simultaneous determination of saikosaponin a and saikosaponin d in Chaihu oral solution by HPLC-DAD method has the advantages of simple operation, good repeatability, high recovery rate, and symmetrical peak shape. It can be used as a Assay method.
