摘 要:目的:探讨枳实薤白桂枝汤对心肌缺血模型兔心电图(ECG)、血管舒缩因子NO和内皮源性内皮素前体(BigET-1)的影响。方法:将50只SPF级家兔随机分为模型组,枳实薤白桂枝汤高、中、低剂量组,地尔硫卓组。经预处理后观察各组家兔注射ET-1后5、10、15、30 min的心电图S-T段与T波的改变,检测各组家兔血管内皮舒张因子NO及BigET-1含量水平的变化。结果:与模型组比较,中、高剂量枳实薤白桂枝汤和地尔硫卓均可使T波回落,使S-T段下降(P<0.05);同时,给药后,枳实薤白桂枝汤中、高剂量组家兔在0.5、2、6 h时,地尔硫卓组在0.5 h时,血管内皮舒张因子NO明显高于模型组,差异有统计学意义(P<0.05);各组家兔BigET-1水平注射ET-1后0.5 h时明显升高,2 h回落到基线水平;注射ET-1后0.5 h,枳实薤白桂枝汤各剂量组、地尔硫卓组BigET-1水平均低于模型组(P<0.05)。结论:枳实薤白桂枝汤可有效地作用于心肌细胞,升高NO,降低BigET-1,对心肌缺血具有一定的保护作用。Objective: To investigate the effects of Zhishi Xiebai Guizhi Decoction on the electrocardiogram(ECG), vasomotor factor NO and endothelin precursor(BigET-1) of rabbits with myocardial ischemia. Methods: A total of 50 SPF rabbits were randomly divided into model group, Zhishi Xiebai Guizhi Decoction high, medium and low-dose group, and diltiazem group. After pretreatment, the changes of S-T segment and T wave of ECG were observed at 5, 10, 15, 30 min after ET-1 injection in each group of rabbits, and the changes in the levels of vascular endothelial dilation factor NO and BigET-1 were detected in each group of rabbits. Results:Compared with the model group, the medium and high-dose of Zhishi Xiebai Guizhi Decoction and diltiazem can reduce the T wave and decrease the S-T segment(P <0.05). At the same time, after administration, the vascular endothelial relaxing factor NO of rabbits in Zhishi Xiebai Guizhi Decoction medium and high-dose group was significantly higher than that of the model group at 0.5, 2, and 6 h, and the vascular endothelial relaxing factor NO in the diltiazem group was significantly higher than the model group at 0.5 h, and the difference was statistically significant(P <0.05). The levels of BigET-1 in rabbits of each group increased significantly at 0.5 h after drug injection, and fell back to the baseline level at 2 h. At 0.5 h after injection of ET-1, the levels of BigET-1 in Zhishi Xiebai Guizhi Decoction high, medium and low-dose group and diltiazem group were lower than those in model group(P<0.05). Conclusion: Zhishi Xiebai Guizhi Decoction can effectively act on myocardial cells, increase NO, reduce BigET-1, which has a certain protective effect on myocardial ischemia.
